Methods for detection of protein-protein interactions in telomere studies

Abstract:

Telomery jsou nukleoproteinové komplexy představující konec chromozomu, mají řadu funkcí, především chrání chromozomy před jejich fúzí a nukleázovou degradací a podílejí se na řízení buněčného cyklu. Jejich délka, struktura a funkce je striktně regulovaná množstvím telomer-asociovaných proteinů včetně telomerázy, jen některé z nich byly dosud popsány a přesný způsob regulace telomer není znám. Jedním z přístupů vedoucích k pochopení této regulace je hledaní nových telomer-asociovaných proteinů interagujících s již charakterizovanými proteiny. K tomuto účelu je využívána např. metoda dvojhybridního systému umožňující jak detekci protein-proteinových interakcí mezi klonovanými proteiny, tak nalezení interagujících proteinů v cDNA knihovně. Kromě této techniky existuje řada dalších metod např. koimunoprecipitace a další techniky in vitro, které tento systém doplňují a používají se k ověření jeho výsledků. Cílem bakalářské práce bude zpracovat přehled v současnosti nejužívanějších postupů pro charakterizaci protein-proteinových interakcí a získat tak teoretický základ pro experimentální práci v rámci navazující diplomové práce.

Abstract:

Telomeres are nucleoprotein complexes at the very end of chromosomes and have several important functions.They protect chromosome from end-to-end fusion and from nuclease degradation and they are also involved in cell cycle regulation. Their lenght, architecture and functions are tightly regulated by telomere asociated proteins, including telomerase enzyme. Till nowadays, only a few of theme were charakterizied and the maintaining and regulatory mechanism of telomeres is not fully clear yet. One of good aproach, how to understand these processes is to make an effort to find and charakterize new proteins interacting with charaterized telomere binding proteins. For this purpose e.g. is employed two-hybrid system – method enabling not only detection of protein-protein interactions between cloned proteins, but also a powerfull method for screening cDNA libaries and searching for new interacting proteins. There are also several other in vivo and in vitro methods, e.g. Co-immunoprecipitation, which is often used to verify data obtained by two-hybrid system. This bacheolor’s work aims on methods for detection and analyzing of protein-protein interactions and gain basics for further diploma studies.
