Occurrence, distribution and management of Buxus pathogens in the Czech Republic

Abstract:

Zimostráz (Buxus spp.) je významnou stálezelenou okrasnou dřevinou, jejíž zdravotní stav je v posledních letech významně ohrožován patogeny r. Calonectria (C. pseudonaviculata a C. henricotiae). V České republice bylo vybráno 27 historických zahrad, kde byl hodnocen jejich zdravotní stav zimostrázu. Na 73 porostech z 27 lokalit byl identifikován patogen r. Pseudonectria (29 vzorků) a patogen r. Calonectria (4 vzorky). Pomocí PCR analýzy byly patogeny r. Calonectria determinovány jako C. pseudonaviculata (1) a C. henricotiae (3). Druh C. henricotiae byl na území České republiky nalezen poprvé. Na dvou lokalitách (Studenec a Žabčice) bylo testováno osm vybraných druhů a kultivarů zimostrázu na náchylnost k patogenu C. henricotiae a ve dvou variantách závlahy (sprejovými postřikovači a kapkovou závlahou). Byla prokázána statisticky významně vyšší náchylnost druhu Buxus sempervirens oproti B. microphylla. U rostlin zavlažovaných kapkovou závlahou prokázána nižší intenzita napadení oproti variantě se sprejovými postřikovači. V laboratorních a polních podmínkách byl testován inhibiční vliv účinných látek na růst mycelia izolovaných kultur r. Calonectria. Nevyšší účinnosti pro všechny 4 izoláty dosahovaly látky tebuconazol + fluopyram, prochloraz, cyprodinil + fludioxonil a thiophanate-methyl. Ideální teplota pro růst mycelia C. pseudonaviculata a C. henricotiae byla stanovena na 24 °C. Při vzájemném křížení izolátů nedošlo k vytvoření pohlavního stadia houby. Byla poprvé testována náchylnost druhů čeledi Buxaceae (Pachysandra a Sarcococca) k patogenu C. henricotiae. Byla popsána epidemiologie patogenu C. henricotiae na druhu Pachysandra ve dvou odlišných mikroklimatických podmínkách. Pro praxi byla navržena metodika preventivních a ochranných opatření zimostrázu proti patogenu Calonectria spp.

Abstract:

Boxwood (Buxus spp.) is an important ornamental evergreen woody plant. Its health is currently being threatened by fungi of the genus Calonectria (C. pseudonaviculata and C. henricotiae). A survey of boxwood plantings (27 historic gardens) was conducted in the Czech Republic. In total, 73 boxwood samples were evaluated, from which the pathogen Pseudonectria was identified on 29 samples and Calonectria on four samples. The samples were determined as C. pseudonaviculata (1) and C. henricotiae (3) by PCR analysis. This is the first report of the species C. henricotiae in the Czech Republic. The susceptibility of eight selected species and cultivars of boxwood to the pathogen C. henricotiae was tested at two locations (Studenec and Žabčice) and in two variants of irrigation (spray sprinklers and drip irrigation). The species Buxus sempervirens and its cultivars were found to be statistically significantly more susceptible to the pathogen C. henricotiae in comparison to the B. microphylla species. The intensity and level of infestation of the disease was significantly lower on plants irrigated with drip irrigation, than by the variant with spray sprinklers.Fungicide efficacy on mycelia growth of isolated Calonectria cultures was evaluated in vitro and in vivo. Active ingredients tebuconazole + fluopyram, prochloraz, cyprodinil + fludioxonil and thiophanate-methyl showed the highest efficacies on all 4 isolates. The optimal temperature of 24 ° C for mycelia growth of C. pseudonaviculata and C. henricotiae was tested. No sexual reproduction was observed, when pairing isolates with themselves.For the first time the susceptibility of other species of the Buxaceae family (Pachysandra and Sarcococca) to the pathogen C. henricotiae was tested. The epidemiology of the C. henricotiae pathogen on Pachysandra sp. in two different microclimatic conditions was described.Methodology of preventive management and boxwood blight treatment was proposed.
