Investigation of the Francisella tularensis glycoproteome

Abstract:

The glycosylation is among the most common post-translational modifications, having an influence on the protein properties and playing a crucial role in many biological processes such as cell-to-cell recognition, cell adhesion or modulating the immune response. It has been found that glycoproteins of many pathogenic bacteria are associated with virulence factors. So far, a presence of glycoproteins has not been studied in a highly infectious Gram-negative bacterium Francisella tularensis, the causative agent of tularemia. The scope of this thesis was to investigate the proteome of Francisella tularensis for the presence of glycosylation as a possible factor of virulence. The present work has demonstrated that the steps leading to the identification of bacterial protein glycosylation can differ significantly from those used for investigation of glycosylation in eukaryotes. Commonly used glycomics approach of the complex protein mixture from eukaryotes is not applicable to bacterial, or at least Francisella, cell lysates. Thus, the glycan structures were eventually determined indirectly, using tools that deals with glycopeptides. In this work, a wide spectrum of putative Francisella tularensis glycoproteins was characterized through the combination of carbohydrate-specific detection and enrichment techniques, on the basis of both hydrazide chemistry and lectin affinity, with highly sensitive mass spectrometry utilizing the bottom-up proteomic approach. Among the putative glycoproteins, glycosyltransferases, immunoreactive proteins, outer membrane proteins, lipoproteins, components of type IV pili and type II secretion system, and Francisella pathogenicity island-encoded proteins were identified. Aforementioned methods for glycoprotein detection and enrichment pointed out 12 proteins, which represent the best candidates for Francisella tularensis glycosylation. Further detail investigation of putative glycoproteins FTH_1071 and FTH_0069 led to the structural characterization of an identical glycan structure modifying both proteins and the identification of glycan-modified peptides. Further definition of the proteins modified by glycan(s) is needed, faciliting a better understanding of the possible function of protein glycosylation in pathogenicity of this microbe.

Glykosylace se řadí mezi nejčastější post-translační modifikace, mající vliv na vlastnosti proteinů a hrající zásadní úlohu v mnoha biologických procesech, jakými jsou mezibuněčné rozpoznávání, buněčná adheze či modulace imunitní odpovědi. Glykoproteiny mnoha patogenních bakterií jsou spojovány s faktory virulence. Přítomnost glykoproteinů dosud nebyla studována ve vysoce virulentní Gram-negativní bakterii Francisella tularensis, původci onemocnění tularémie. Cílem této práce bylo ověřit, zda se v proteomu Francisella tularensis vyskytuje glykosylace, jakožto pravděpodobný faktor virulence. Předkládaná práce poukazuje na skutečnost, že kroky vedoucí k identifikaci glykosylace bakteriálních proteinů se významně odlišují od kroků používaných v analýze glykosylací eukaryot. Běžně používané glykomické studie komplexních směsí proteinů eukaryotického původu nelze použít v případě analýzy bakteriálních buněčných lyzátů, nebo alespoň ne v případě bakterie Francisella tularensis. Glykanové struktury proto byly určeny nepřímo, a to s použitím nástrojů studujících glykopeptidy. V této práci bylo prostřednictvím tzv. "bottom-up" proteomického přístupu charakterizováno široké spektrum potenciálních glykoproteinů Francisella tularensis. Využito přitom bylo kombinace sacharid-specifických detekčních a obohacovacích technik, založených na hydrazidové chemii a lektinové afinitě, s vysoce citlivou hmotnostní spektrometrií. Mezi identifikovanými potenciálními glykoproteiny byly glykosyltransferázy, imunoreaktivní proteiny, zevní membránové proteiny, lipoproteiny, složky sekrečních systémů typu IV a II a proteiny kódované geny z ostrova patogenicity. Výše zmíněné metody pro detekci a obohacení glykoproteinů poukázaly na 12 proteinů, které představují nejlepší kandidáty pro glykosylaci proteinů Francisella tularensis. Následné detailní studium potenciálních glykoproteinů FTH_1071 a FTH_0069 vedlo k charakterizaci totožné glykanové struktury modifikující oba proteiny a k identifikaci peptidů modifikovaných tímto glykanem. Pro lepší porozumnění pravděpodobné funkce glykosylace proteinů v patogenitě tohoto mikroba je vhodné, aby byly popsány další proteiny modifikované glykosylací.
