Abstract:

Abstrakt Tato disertace se zabývá aplikacemi a vývojem metod kapilární zónové elektroforézy pro analytiku přírodních produktů extrahovaných z rostlin jako je Huperzia Serrata (Huperzine A) a některými sloučeninami (polyfenoly) z šalvěje (Salvia Officinalis). Použití metod umělých neuronových sítí v kombinaci s metodou plánování pokusů je rovněž zahrnuto v této disertaci. Nové analytické postupy pro stanovení Huperzinu A ve vodných roztocích a v krevním séru byly vyvinuty za použití umělých neuronových sítí. V druhé části práce byly pak detekční limity podstatně zlepšeny a nově vypracovaný postup umožňuje detekci tohoto analytu s detekčním limitem o 2 řády nižším než při UV detekci. Tato metoda je založena na nové, dosud nepopsané reakci Huperzinu A s RBITC činidlem, reakcí pro tento typ aminu dosud nepopsanou v literatuře. Po derivatizaci Huperzinu A tímto činidlem je produkt reakce, jehož struktura byla prokázána MALDI TOF hmotnostní spektrometrií, detekován metodou laserem indukované fluorescence (LIF). Byla rovněž vyvinuta nova analytická metoda kapilární zónové elektroforézy pro separaci šesti strukturně podobných látek jako je epicatechin, catechin, kyselina vanilinová, rozmarýnová, kávová, a kyselina gallová z extraktu šalvěje ( Salvia officinalis). Jednotlivé látky byly identifikovány metodou přídavků standardů těchto sloučenin k analyzovaným extraktům. Rovněž v této části Disertace byla metodologie umělých neuronových sítí v kombinaci s plánováním pokusů užita pro optimalisaci separací a optimalisaci metody. Závěrem, několik nových analytických postupů či metod bylo v této práci vyvinuto a validováno.

Abstract:

Abstract This thesis deals with the use of the Capillary electrophoresis technique for the analysis of natural products that are extracted from the plant Huperzia Serrata (Huperzine A) and some compounds (polyphenols) from Salvia Officinalis. The use of artificial neural networks in combination with experimental design for the optimization is included in this Thesis, as well. The new CE method for the determinations of Huperzine A was developed using artificial neural networks in aqueous and serum samples. In the second part of the Thesis, the limit of detection was improved substantially and new approach enabled the detection of the analyte two orders of magnitude lower than the limit of detection that is achievable by UV detection. The method is based on a new reaction of Huperzine A with suitable reagent (RBITC), which was not described in the literature before. The structure of the product was determined bz MALDI TOF mass spectrometry. The reagent was used for the derivatization of HuperzineA and the product was detected by laser induced fluorescence (LIF). A new Capillary Electrophoresis (CE) method was developed for the separation of six structurally similar compounds such as epicatechin, catechin, vanillic, rosmarinic, caffeic, and gallic acids from Salvia officinalis plants. The analytes were distinguished by spiking a standard dose of polyphenols to the samples. ED-ANNs approach was also used here in order to optimise the separation. Concluding, several new analytical methods were developed and validated.
