Expression and role of LGR5 in development of craniofacial structures

Abstract:

Vývoj kraniofaciální oblasti je komplikovaný proces, na kterém se podílejí buňky neurální lišty. Tyto buňky migrují, diferencují a dávají vzniknout řadě kraniofaciálních struktur, jako například patro, vomeronasální orgán či zuby. Prenatální i postnatální vývoj kraniofaciálních struktur je určen kmenovými a progenitorovými buňkami. Tyto buňky se dělí, proliferují a také diferencují do příslušných buněčných typů. Přetrvávání těchto buněk až do dospělosti umožňuje sebeobnovu tkání, která je důležitá zejména pro jejich reparaci. Tato práce se zabývá detekcí progenitorových buněk v průběhu vývoje kraniofaciálních struktur pomocí markeru LGR5. Jedná se o transmembránový receptor, který byl identifikován jako marker dospělých kmenových buněk v mnoha tkáních. Během našeho výzkumu jsme odebírali embrya transgenních myších linií LGR5-EGFP-IRES-creERT2 v různých stádiích embryonálního a postnatálního vývoje. Pomocí imunofluorescenčního barvení byla detekována exprese LGR5 zejména v oblasti mezenchymu přiléhajícího k nosnímu septu, v mezenchymu na labiální straně záhybu podél jazyka, v mezenchymu kolem nesenzorického i senzorického epitelu vomeronasálního orgánu, v mezenchymu labiální strany zubní stopky či v epitelu na linguální straně této stopky. Díky detekci koexprese LGR5 s dalšími markery (SOX2, SOX9 a FZD6) bylo zjištěno, že k překryvu exprese LGR5 se SOX2, SOX9 i FZD6 dochází pouze v oblasti epitelu linguální strany zubní stopky. Osud buněk byl sledován zejména v průběhu odontogeneze s využitím embryí Rosa26-lacZ reportérových myší, a to ve třech časových intervalech. Po odebrání embryí byla pomocí LacZ barvení odhalena přítomnost LGR5/LacZ-pozitivních buněk jen v několika oblastech vyvíjejícího se zubu, jako je zubní stopka, náhradní zubní lišta či vnitřní sklovinný epitel. V rámci jiných kraniofaciálních struktur byly LGR5/LacZ-pozitivní buňky nalezeny v mezenchymu podél záhybu jazyka či v oblasti temporomandibulárního kloubu. Díky těmto analýzám bylo zjištěno, že ačkoliv byl dosud LGR5 popsán zejména jako epitelový marker progenitorových buněk, během vývoje kraniofaciální oblasti byl odhalen i v řadě populací mezenchymových buněk. Koexprese s dalšími markery progenitorových buněk (SOX2, SOX9) odhalila existenci nové dosud nepopsané niky progenitorových buněk v zubní stopce molárů. Díky mapování osudu LGR5-pozitivních buněk bylo zjištěno, že se tyto buňky výrazně nepodílí na formování zubního základu, ale spíše dávají vzniknout rudimentální zubní liště.

Abstract:

Development of vertebrate head structures comprises of an intricate and complex series of molecular and celullar events which are strictly controled. Migration and differentiation of cranial neural crest cells is lagerly responsible for the formation .of craniofacial structures, such as palate, vomeronasal organ and teeth. Both prenatal and postnatal craniofacial development is driven by various population of stem/progenitor cells. They proliferate and differentiate into specific cell types during embryonic development and also play a role in adult tissue repair due to their self-renewal capacity. The goal of this thesis is the identification and characterization of LGR5 positive progenitor cells in craniofacial region. LGR5 is a transmembrane protein receptor, which has been established as a marker of adult stem cells in certain types of tissues. We collected several embryonic and postnatal stages of transgenic mouse lines LGR5-EGFP-IRES-creERT2. Expression of LGR5 was detected by imunofluorescence in the area of mesenchyme adjacent to nasal septum, in the mesenchyme of labial fold along the tongue, in the mesenchyme surrounding both non-sensory and sensory epithelium of vomeronasal organ, in the mesenchyme of labial side and in the epithelium lingual side of the dental stalk. Co-expression analysis of LGR5 with other progenitor markers (SOX2, SOX9 a FZD6) revealed that protein expression of LGR5 and SOX2, SOX9 and FZD6 overlaped only in the epithelium of lingual side of the dental stalk. Lineage tracing of the LGR5-positive cells was investigated particularly during odontogenesis using Rosa26-lacZ reporter mice at three time points. LacZ staining uncovered small population of LGR5 /LacZ positive cells in few areas of developing tooth, including dental stalk, succesional dental lamina or inner enamel epithelium. LGR5 /LacZ positive cells were also detected in other craniofacial structures such as mesenchyme of lateral lingual fold and temporomandibular joint. Up to now, LGR5 has been characterized as a marker of epithelial progenitor cells, however, our study revealed its expression also in the large range of mesenchymal cells populations during craniofacial development and demonstrated the presence of a novel niche of progenitors in molar dental stalk, which has not been described yet. Lineage tracing of LGR5-positive cells indicated that these cells are rather involved in rudimental dental lamina than in forming of tooth germ.
