Potential targets for vaccine development in pathogenic treponemes

Abstract:

V bakalářské práci se věnuji onemocnění syfilis a charakterizuji jejího původce Treponema pallidum subsp. pallidum spolu s dalšími patogenními treponemami. Dále se zaměřuji na hledání možných kandidátů pro vývoj vakcíny proti syfilis. Celkem popisuji výsledky imunizace prostřednictvím 21 rozdílných struktur. Mezi tyto struktury patří především proteiny s antigenní povahou a povrchovou lokalizací, které tvoří většinovou část kandidátů. Dále byly pro účely imunizace testovány i celé bakterie či protilátky a T-lymfocyty. Průběh infekce, resp. účinnost vakcíny je hodnocena podle povahy lézí, rychlosti jejich vzniku a pozitivity na přítomnost živých motilních treponem. Většinou bylo dosaženo jen částečné protekce, kvůli čemuž hrozí, že infekce bude asymptomatická, zůstane nerozpoznána a vyvine se v pozdější stádia. V ojedinělých případech imunizace spíše zhoršila průběh infekce. Vakcinací se plné protekce dosáhlo jen v jediném případě celobuněčné vakcíny, nicméně vakcinační protokol byl příliš náročný, než aby mohl být zaveden do praxe.

Abstract:

In this thesis I deal with disease syphilis and characterizate its causative agent Treponema pallidum subs. pallidum together with anothers patogenic treponemes. I searched also for potential candidates for the development of vaccine against syphilis. In total, this thesis describes the results of immunization with 21 different structures, including antibodies and T-lymphocytes used in passive immunization, whole cells, and proteins with antigenic characer and surface localization, which form the majority of candidates. The efficiency of the vaccine is determined by the nature of lesions, time of their appearence and positivity for the presence of live motile treponemes. Only partial protection is usually achieved, which might lead to asymptomatic infection and development of later stages of disease. In several cases, the immunization rather worsened the course of infection. Full protection by vaccination was reached in only one case, using a whole-cell vaccine, but the vaccination protocol was too demanding to be applied in practice.
