Abstract:

V poslední době se zvyšuje zájem o nanotechnologie, zejména o liposomy a mikrobubliny, které nacházejí - díky svým výjimečným vlastnostem - využití jak v diagnostice, tak i v terapii. I přes značnou podobnost se liší mikrobubliny od liposomů hlavně velikostí, která je u mikrobublin v rozmezí mikrometrů. To může být praktické při klinickém použití, protože mikrobubliny mají podobnou velikost jako erytrocyty. Mikrobubliny jsou obvykle složeny z biokompatibilních látek, jako je cholesterol, fosfolipidy a lipidy, díky čemuž je málo pravděpodobné, že by mohly při aplikaci vyvolat imunogenní reakci. Kromě toho je možné obal mikrobublin různě modifikovat a využít je tak jako nosiče léčiv a genů. Hlavním cílem mé disertační práce "Mikro a nanopartikulární částice jako nosiče léků: Povrchově modifikované mikrobubliny využívané jako ultrazvukové kontrastní látky a nosiče léčiv" je studium přípravy, fyzikálně-chemická charakterizace, diagnostické jakož i terapeutické využití mikro a nanočástic, a to zejména mikrobublin. První kapitola je věnována základním principům zobrazovací techniky ultrazvuku, který je klíčovým faktorem při klinickém použití mikrobublin. Další část je zaměřena na obecnou charakteristiku mikrobublin a jejich bezpečnost v humánní klinické diagnostice, zatímco třetí kapitola se zabývá praktickou aplikací mikrobublin v medicíne a výzkumem za pomoci ultrazvuku jako základního zobrazovacího zařízení, a to zejména se zaměřením na chování mikrobublin v ultrazvukovém poli in vivo. Třetí kapitola poskytuje zároveň stručný přehled možných modifikací povrchu mikrobublin, což má velký význam při využití mikrobublin jako nosičů léčiv či genů, nebo při snaze o rozbití krevních sraženin, čemuž jsou ve třetí kapitole věnovány příslušné podkapitoly. Čtvrtá kapitola popisuje různé metody přípravy mikrobublinjako jsou například sonikace, mechanické třepání, membránová emulsifikace a využití mikrofluidizérů. Pátá i šestá kapitola je založena na metodách, které jsem využíval při své experimentální práci, a na výsledcích experimentů, které byly následně publikovány v impaktovaných časopisech. Použité metody a postupy pokrývají problematiku přípravy mikrobublin, jejich fyzikální a chemickou charakterizaci, jakož i jejich uchování pomocí lyofilizace. Tyto výsledky jsou níže diskutovány a je nastíněn možný směr dalšího vývoje v oblasti diagnostických mikrobublin a zařízení sloužících k prodloužení cirkulace mikrobublin v krevním řečišti.

Abstract:

Recently, there has been a strong upsurge of interest in the field of nanotechnologies, represented particularly by liposomes and microbubbles, which have become - due to their exceptional characteristics - vastly used both in diagnostics and therapy. Despite the very similar design, microbubbles differ from liposomes crucially. Due to their micrometre size, they can be very practical in clinical use. In organisms, they significantly resemble erythrocytes and therefore they behave very much alike. Being usually composed of physiological components such as cholesterol, lipids, and phospholipids, they are also very unlikely to cause any immunogenic reaction when applied in organisms. Moreover, a variety of structures can be attached to the microbubble surface, making them especially suitable for drug and gene delivery. The principal target of my dissertation thesis entitled “Micro and Nanoparticles as Drug Carriers: Surface-Modified Microbubbles Used as Ultrasound Contrast Agents and Drug Carriers” is the study of preparation, physico-chemical characterisation and diagnostic as well as therapeutic application of micro and nanoparticles, particularly microbubbles. The first chapter is devoted to elementary principles of ultrasound imaging techniques, which is the pivotal factor in clinical use of microbubbles. The next part focuses on the general characterisation of microbubbles and their safety in human clinical diagnostics, while the third chapter is concerned with the practical applications of microbubbles in medicine and research with using ultrasound as a basic imaging device, especially focusing on the behaviour of the microbubbles in the ultrasound field in vivo. The third section also provides a brief overview of the possible modification of the surface of microbubbles, which is very important in the use of microbubbles as carriers of drugs or genes, or in an attempt to break up blood clots what is in the third chapter devoted to the relevant chapters. The fourth chapter describes the different methods of preparation of microbubbles such as sonication, mechanical agitation, membrane emulsification or the use of microfluidic devices. Both the fifth and sixth chapter are based on the methodologies and results of experiments that were carried out in my experimental work and subsequently published in journals. The used methods and procedures cover the field of preparation of microbubbles, their physical and chemical characterisation, and their preservation by lyophilisation. Finally, these results are discussed and possible directions for further developments in the field of diagnostic microbubbles and devices used to extend the circulation of microbubbles in the bloodstream are outlined.
