Involvement of circular RNAs in pathogenesis of glioblastomu

Abstract:

Multiformní glioblastom (GBM) je vysoce maligní primární intrakraniální nádor s průměrnou dobou celkového přežití pacientů od 12 do 15 měsíců po stanovení diagnózy. I přes současnou konvenční léčbu se prognóza pacientů s GBM prakticky nijak nezlepšuje. Důvodem je vysoká heterogenita nádoru, jeho značná chemoradiorezistence a s tím související časné recidivy onemocnění. Jsou však intenzivně zkoumány nové možnosti terapie, a především nové prognostické a prediktivní biomarkery odpovědi na léčbu. Od objevu nekódujících RNA a jejich regulačního potenciálu na mnoha úrovních genové exprese se velká část výzkumu GBM zaměřuje právě na tyto molekuly. Relativně nedávno objevenou skupinou nekódujících RNA jsou cirkulární RNA (circRNA). Jejich hlavní doposud popsanou úlohou je schopnost vázat specifické mikroRNA (miRNA) a případně je následně opět uvolňovat. Zajišťují tak v buňce dostupnost těchto krátkých RNA v čase. Klíčová úloha miRNA u nádorových onemocnění včetně GBM již byla mnohokrát ověřena. Není proto překvapující, že se již objevily studie popisující aberantní expresi některých circRNA v GBM a schopnost těchto cirkulárních molekul vázat právě miRNA hrající v biologii GBM důležitou roli. Detailnější studium circRNA u GBM by tak mohlo odhalit nejen klinicky využitelné prediktivní a prognostické biomarkery, ale také nové efektivní terapeutické cíle. Hlavními cíli diplomové práce bylo za pomoci sekvenování nové generace identifikovat konkrétní molekuly circRNA v GBM a nenádorové mozkové tkáni, pomocí metody qRT-PCR validovat expresi circRNA vybraných na základě recentní odborné literatury v souboru nádorových i nenádorových vzorků a v neposlední řadě in vitro s využitím metody „pull-down assay“ verifikovat schopnost těchto circRNA vázat predikované miRNA. Výsledky této práce představují pilotní data, která poslouží jako podklad pro přípravu tuzemských i zahraničních grantových žádostí.

Abstract:

Glioblastoma multiforme (GBM) is a highly malignant primary intracaranial tumour with median overall survival of patients ranging from 12 to 15 months after the initial diagnosis. Despite currently available conventional treatment, the prognosis for patients with GBM is unfortunately still very poor. The primary reason for this is often thought to be extensive heterogeneity of the tumour, its chemoresistance, and, related to that, an early occurrence of relapse. New ways of treatment are being studied intensively with research being focused on therapeutic targets and prognostic and predictive biomarkers of response to treatment. Since the discovery of non-coding RNAs and their regulatory potential at various levels of gene expression, much of the research into GBM has been focused on these molecules. A relatively recently discovered group of non-coding RNAs are circular RNAs (circRNAs). Their main currently described role is the ability to bind specific microRNAs (miRNAs) and release them again if needed. This ensures the availability of these short RNAs over time in the cell. The key role of miRNAs in cancer, including GBM, has been proven many times. Therefore, it is not surprising that some studies describing the aberrant expression of some circRNAs in GBM and the ability of these circular molecules to bind miRNAs and affect their function in GBM biology have already been published. More detailed study of circRNAs in GBM could reveal not only clinically useful predictive and prognostic biomarkers but also new effective therapeutic targets. The main aim of this thesis was to identify particular circRNA molecules in GBM and non-tumor brain tissue using new generation sequencing, validate the expression of circRNAs selected on the basis of recent scientific literature in the set of tumor and non-tumor samples using qRT-PCR and, last but not least, verify the ability of circRNAs to bind predicted miRNAs using RNA pull-down assay. Results presented in this thesis are pilot data which will serve as a basis for the preparation of domestic and foreign grant applications.
