Effect of growth phase of winter wheat on the activation of COR/LEA genes under simultaneous exposure to low temperatures

Abstract:

Tato bakalářská práce se zaměřuje na kvantifikaci exprese vernalizačních a ochranných genů zapojených do procesu chladové aklimatizace a na zjištění jejich možné regulace, u pšenice ozimé. Z polních podmínek byly odebrány vzorky listů a růstových vrcholů z odrůdy Bohemia a RGT Sacramento. Ze vzorků byla izolována RNA, která byla pomocí reverzní transkripce převedena na cDNA a následně byla kvantifikována pomocí metody qPCR. Z výsledků byla vypočítána normalizovaná relativní exprese každého měřeného genu. Podle výsledků měl přechod z vegetativní do generativní fáze za následek zvýšení exprese vernalizačních genů a ve většině případů snížení exprese genů chladové aklimatizace. Korelační analýza prokázala negativní korelaci genu VRN-1 a genu WCOR410, WCOR14 a WDHN13 před vernalizací. Po vernalizaci byla prokázána jediná negativní korelace genu VRN-1 a WCOR410.

Abstract:

This bachelor thesis focuses on the quantification of the expression of vernalization and protection genes involved in the process of cold acclimatization and their possible regulation in winter wheat. Leaf and growth apex samples were collected from field conditions from the cultivars Bohemia and RGT Sacramento. RNA was isolated from the samples, converted to cDNA by reverse transcription and then quantified by qPCR. From the results, the normalized relative expression of each gene measured was calculated. According to the results, the transition from vegetative to generative phase resulted in an increase in the expression of vernalization genes and in most cases a decrease in the expression of cold acclimation genes. Correlation analysis showed a negative correlation between VRN-1 and WCOR410, WCOR14 and WDHN13 genes before vernalization. After vernalization, a single negative correlation between the VRN-1 gene and WCOR410 was demonstrated.
