Immobilized recombinant hyaluronan lyase (SpHyl) - preparation of the carrier for the fragmentation pre-column

Abstract:

Diplomová práce se zabývá imobilizací rekombinantní hyaluron lyázy (SpHyl) na vybrané magnetické částice. Cílem práce je vyhodnotit účinnost imobilizace, stanovit operační stabilitu a aktivitu enzymů imobilizovaných na těchto částicích. Práce se zaměřuje na přípravu nosiče pro předseparační HPLC kolonu. Teoretická část práce popisuje strukturu, vlastnosti a významu kyseliny hyaluronové. Dále enzymy štěpící kyselinu hyaluronovou a možnosti jejich imobilizace na magnetické částice. Experimentální část zahrnuje proces imobilizace SpHyl na vybrané částice, ověření její účinnosti pomocí gelové elektroforézy a následné spektrofotometrické stanovení operační stability a aktivity solubilního a imobilizovaného enzymu, stejně jako stanovení kinetických parametrů pro charakterizaci připravených nosičů. Výsledky poskytují poznatky o vhodnosti použitých částic pro předseparační kolonu připravenou pro firmu Contipro a.s., kde budou v nejbližší době prováděny aplikační experimenty.

Abstract:

The thesis deals with the immobilization of recombinant hyaluronidase (SpHyl) on selected magnetic particles. The aim of the thesis is to evaluate the efficiency of immobilization, determine the operational stability, and activity of enzymes immobilized on these particles. The work focuses on the preparation of a carrier for a preseparation HPLC column. The theoretical part of the thesis describes the structure, properties, and significance of hyaluronic acid. Furthermore, it discusses enzymes cleaving hyaluronic acid and the possibilities of their immobilization on magnetic particles. The experimental part includes the process of immobilizing SpHyl on selected particles, verifying its efficiency using gel electrophoresis, and subsequent spectrophotometric determination of operational stability and activity of soluble and immobilized enzymes, as well as determination of kinetic parameters for the characterization of prepared carriers. The results provide insights into the suitability of the particles used for the preseparation column prepared for the company Contipro a.s., where application experiments will be conducted in the near future.
