Effects of changes in xylem sap composition on the plant transpiration rate

Abstract:

V rostlině dochází při stresu z nedostatku vody ke zpomalení transpirace i růstu, a tyto změny jsou regulovány pomocí dálkových signálů. V dnešní době je známo, že se nejedná o izolované signály, ale spíše o jejich síť. Cílem mé diplomové práce bylo otestovat vliv vybraných potenciálních signálů a doplnit tak poznatky o mechanizmech regulujících transpiraci rostlin za sucha. Testovala jsem samostatné působení citrátu, malátu a sulfátu a jejich vliv na transpiraci v kombinaci s pH a kyselinou abscisovou (ABA). Efekt sledovaných látek jsem měřila u odřezaných listů slunečnice, řepky, ředkvičky, pšenice a hrachu a ke stanovení průběhu transpirace v čase jsem využívala gravimetrickou metodu. Sledované látky jsem listům dodávala ve formě umělé xylémové šťávy, do které jsem ponořila řapíky odřezaných listů. Nejvýznamnější změny jsem naměřila u variant s malátem v kombinaci s pH. Při pH 6,5 rychlost transpirace variant s 0,5mM a 2mM malátem oproti kontrole průkazně poklesla u slunečnice, pšenice a řepky; při pH 7,5 naopak rychlost transpirace stoupala. Při pH 6,5 klesala transpirace slunečnice a pšenice u varianty s 2mM citrátem, při vyšším pH nebyl vliv citrátu prokázán. K interakci citrátu a malátu s ABA nedocházelo. Vliv sulfátu na transpiraci nebyl prokázán samostatně ani v kombinaci s ABA. Jako nejvhodnější z testovaných druhů pro tento experiment se ukázala slunečnice a hrách, nejméně vhodné byly rostliny ředkvičky kvůli své vysoké citlivosti na změny vodního potenciálu.

Abstract:

In plants under water stress decreases of the transpiration rate and reduction in growth occur. These changes are controlled by long-distance signals. Nowadays, it is known that they are not isolated signals, but rather a network of signals. The aim of this thesis was to test the effect of selected potential signals and to expand knowledge of the mechanisms regulating transpiration of plants under drought. I have tested the separate effects of citrate, malate and sulfate, and their effect on transpiration in combination with pH and abscisic acid (ABA). The effect of the substances was measured in cutted leaves of sunflower, rape, radish, wheat and pea plants. For stating transpiration rate changes in time the gravimetric method was used. The monitored compounds were supplied to plants in the form of artificial xylem sap into which petioles of the cutted leaf were immersed. I have detected the most pronounced changes in the experiments with malate in combination with pH. In experimental variants with 0.5mM and 2mM malate at pH 6.5 the transpiration rate significantly decreased (compared to the control) in sunflower, wheat and rape, on the contrary, at the pH 7.5 the transpiration rate increased. In variants with 2mM citrate at the pH 6.5 the transpiration rate decreased in sunflower and wheat. At higher pH values no significant effect of citrate has been proven. Also no interaction between citrate and malate has been showed. The influence of sulfate on transpiration rate has not been proved neither separately nor in combination with ABA. Of the tested species sunflower and pea plants proved to be the most suitable for this experiment, conversely the experiment has showed radish to be less suitable due to its high sensitivity to water potential changes.
