Abstract:

Tato disertační práce se zabývá zhášením luminiscence vybraných molekul ve vodných rozpouštědlech a jeho následným potlačením s využitím těžké vody. Luminiscence některých látek může být zhášena molekulami vody přítomnými v rozpouštědle. Na základě měření bylo navrženo několik mechanismů, které by mohly být zodpovědné za snižování intenzity luminiscence v H2O. Pokud nahradíme lehkou vodu (H2O) těžkou (D2O), dochází u některých látek k prodloužení střední doby života a nárůstu intenzity emitovaného záření. Tento jev je využitelný v analytické chemii pro stanovení obsahu deuterované vody v normální vodě a také analytických technikách spojených s fluorescenční detekcí jako např. kapalinová chromatografie (HPLC), kapilární elektroforéza (CE-LIF), fluorescenční mikroskopie, průtoková cytometrie a další. V rámci disertační práce byly zkoumány různé skupiny analytů (fluorescenční značky, proteiny, léčiva, sondy apod.), z nichž některé vykazovaly zvýšení intenzity fluorescence o více než 300 %. Tím je možné dosáhnout nižších limitů detekce, snížení spotřeby vzorku a u mikroskopických metod získat lepší rozlišení a zároveň snížit spotřebu značek a sond.

Abstract:

This dissertation thesis deals with the luminescence quenching of chosen luminescent compounds in the water-based solvents and its suppression by heavy water. The source of the luminescence quenching can be mainly the molecules of water present in the solvent. Considering the structure of the fluorophore there can be observed many mechanisms which can be responsible for the decrease of the fluorescence intensity in the light water. A simple exchange of the light water (H2O) for the heavy water (D2O) can generate a longer lifetime and higher fluorescence intensity of fluorophores. This effect can be practical in the field of analytical chemistry for the determination of deuterated water in normal water and vice versa by fluorescence spectroscopy and analytical techniques coupled to fluorescence detection such as liquid chromatography (HPLC), capillary electrophoresis (CE-LIF), fluorescence microscopy, flow cytometry, etc. In this dissertation thesis, the fluorescence behavior of different groups of fluorophores (fluorescent probes and labels, proteins, drugs, etc.) in H2O and D2O was investigated. The observed fluorescence intensity increasement or lifetime prolongation was up to 300 % for some of the chosen fluorophores. This phenomenon allows to achieve lower detection limits, reduction of the sample consumption in some analytical techniques, and decrease the concentration of fluorescent probes and labels or increase the resolution in the field of the microscopic methods.
