Effect of phagocytosis and other non-specific immune system responses on Borrelia burgdorferi sensu lato

Abstract:

Tato bakalářská práce se věnuje reakci mechanismů nespecifické imunity při eliminaci bakterií komplexu Borrelia burgdorferi sensu lato, způsobující onemocnění lymeská borrelióza. Látkové i buněčné mechanismy nespecifické imunity jsou stěžejní pro průběh onemocnění a zodpovědné za vznik zánětu. Z uvedených mechanismů se tato práce věnuje především fagocytóze borrelií a buňkám v tomto procesu zahrnutých, jako jsou makrofágy a neutrofily. Mechanismus fagocytózy namířený proti borreliím vykazuje některé specifické znaky, jako např. odlišný způsob pohlcení a indukce uvolňování určitých cytokinů. Borrelie nejsou citlivé vůči všem likvidačním mechanismům fagocytózy. Pouze některé z nich jsou schopny patogen účinně zlikvidovat. To vše souvisí se vznikem rezistenčních mechanismů, které byly patogenem vyvinuty jako obrana proti imunitním mechanismům hostitele. Z toho důvodu dochází ke komplikované léčbě některých pacientů, nacházejících se v II. a III. stádiu onemocnění. Rešerše této bakalářské práce by mohla napomoci k řešení této problematiky.

Abstract:

This bachelor thesis deals with the reaction of non-specific immunity mechanisms in the elimination of Borrelia burgdorferi sensu lato causing Lyme borreliosis. Non-specific immune and cellular mechanisms are crucial to disease progression and responsible for inflammation. From the above mentioned mechanisms, this work is mainly concerned with phagocytosis of borreliae and cells involved in this process, such as macrophages and neutrophils. The mechanism of phagocytosis directed against borreliae exhibits some specific features, such as a different pattern of uptake and induction of release of certain cytokines. Borrelia is not sensitive to all phagocytosis destruction mechanisms. Only some of them are able to effectively destroy the pathogen. All this is related to the emergence of resistance mechanisms that have been developed by the pathogen as a defense against host immune mechanisms. Therefore, there is a complicated treatment of some patients in II. and III. disease stage. The research of this bachelor thesis could help to solve this problem.
