45S ribosomal DNA organization (rDNA) and 5S rDNA clusters were investigated in the genome of selected representatives of different lineages of algae using PCR and Southern hybridization. Four methods of genomic DNA isolation were tested in order to identify the best method for different groups of algae. V této bakalářské práci byly zkoumány organizace 45S ribozomální DNA (rDNA) a 5S rDNA klastrů v genomu vybraných zástupců jednotlivých vývojových linií řas pomocí PCR a Southern hybridizace. Dále byly testovány 4 metody izolace genomové DNA s cílem nalézt nejvhodnější postup pro jednotlivé skupiny řas.
