Preparation and characterization of pluripotent stem cell-derived cardiomyocyte - In vitro model of cardiomyopathy

Abstract:

Kyselina retinová (retinoic acid, RA) je metabolit retinolu, vitamínu A, působící jako morfogen a teratogen. Ukázalo se, že RA má komplexní účinky na vývoj srdce a WNT signální dráhu, ale vztah mezi nimi stále není plně pochopen. Bylo pozorováno, že během in vitro diferenciace myších embryonálních kmenových buněk může přidání RA nebo inhibice WNT indukovat kardiomyogenezi a následnou buněčnou specifikaci. Experimenty s hiPSCs prokázaly, že spolupráce těchto dvou faktorů hraje klíčovou roli při tvorbě kardiomyocytů. Nicméně, mechanismy jejich regulace stále nejsou zcela pochopeny. Tato práce se zaměřuje na možnou interakci mezi zmíněnou RA a XAV939, WNT inhibitor, a jejich vliv na vývoj buněk srdce.

Abstract:

Retinoic acid (RA) is a metabolite of retinol, vitamin A, acting as morphogen and tera-togen. RA has been shown to have complex effects on heart development and WNT signalling, but the relationship between the two is still not fully understood. We have observed that during in vitro differentiation of murine embryonic stem cells addition of RA or WNT inhibition can induce cardiomyogenesis and subsequently cell specification. Experiments with hiPSCs proved that the cooperation between those two factors play a key role in cardiomyocyte formation. However, mechanisms of their regulation are still not absolutely understood. This thesis focuses on the possi-ble interaction on mentioned RA and XAV939, a WNT inhibitor and their effect on cardiac cell development.
