Monitoring stability of enzymes and enzyme mixtures for the determination of alcohol and glycerol in wine

Abstract:

Bioelektronický jazyk je často popisován jako zařízení, které je schopno komplexního vyhodnocení analytu, je konstruován jako soubor mnoha biosenzorů s odlišnými funkcemi. Tato bakalářská práce je zaměřena na tvorbu biosenzorů, které by se daly využít ke konstrukci bioelektronického jazyka. Obsahem této práce, experimentálního charakteru, je základní popis chemického složení vína a jeho účinků na lidský organismus. Nejdůležitějšími látkami jsou však pro nás ethanol, methanol a glycerol. Následují základní charakteristiky amperometrických enzymatických biosenzorů a výsledky, dle experimentů, nejvhodnějších typů biosenzorů, které poskytovaly dostatečnou – kvantifikovatelnou odezvu. Okrajovou částí je měření stability enzymů v homogenním a heterogenním prostředí.

Abstract:

The bioelectronic tongue, often described as a device that is capable of complex evaluation of analyte, is designed as a set of many biosensors with different functions. This thesis concentrates on the formation of the biosensors that can be used for the construction of the bioelectronic tongue. Content of the thesis, which has an experimental character, is a basic description of a chemical composition of wine and its effects on the human organism. Nevertheless, the most important substances here are ethanol, methanol and glycerol. In the next part, the thesis deals with basic characteristics of amperometric enzymatic biosensors and their most appropriate types, capable of providing a sufficient - qualifiable response, according to the results of the experiments. The thesis is also sketching the basics of measuring of enzymes stability in the homogneous and the heterogeneous environment.
