Mapping of elemental distribution in parts of plants by laser spectrometry

Abstract:

Předmětem této disertační práce je studium distribuce kovů a kovových nanočástic v prostředí, převážně pak v rostlinách, jakožto důležité složce životního prostředí. Většina studií je soustředěna na využití techniky LIBS (spektroskopie laserem buzeného plazmatu) pro stanovení prostorové distribuce kovů a pro rychlou detekci prvků v rostlinách a s tím spjatým vývojem metodiky a instrumentace. Rostliny jsou významnou součástí toku látek a energií v životním prostředí a hrají důležitou roli v mobilizaci, transportu a akumulaci kovů. Zároveň jsou využívány v mnoha současných biotechnologických procesech, jako např. při výrobě biopaliv. Za použití dvoupulzního systému LIBS v orthogonálním uspořádání byly získány mapy prostorového rozložení prvků, konkrétně Cu, Ca, Ag a stříbrných nanočástic, na příčných řezech nadzemních i podzemních orgánů rostlin. Byla nalezena optimální metoda přípravy řezů, při níž bylo dosaženo rozlišení až 50 µm s citlivostí měření na úrovni blízké přirozenému výskytu těchto prvků v rostlinách. Soustava LIBS v dvoupulzním uspořádání byla rovněž využita k rychlé detekci kovů v biofilmech řas a řasových suspenzích. Další část práce je věnována stanovení obsahu prvků v listech a stoncích vrby polární (Salix polaris). Rovněž byla provedena analýza podloží na místech vzorkovaných rostlin. Vzorky byly odebrány ve městě Pyramiden, bývalém těžebním dolu na uhlí, ležícím na arktickém souostroví Špicberky. Bylo provedeno zhodnocení zatížení této oblasti těžkými kovy jako důsledek osídlení a těžby v jinak minimálně dotčené krajině Arktidy.

Abstract:

The aim of the work presented in this thesis is the study of metals and metal nanoparticles in the environment, mostly in plants as the important component of the environment. Most studies are focused on the use of a LIBS method (Laser-induced Breakdown Spectroscopy) for the determination of spatial distribution of metals and for rapid detection of metals in plant samples and the related development of methodology and LIBS instrumentation. Plants are the part of the flow of matter and energy, influence the mobility and play important role in mobilization, transport and accumulation of metals in the environment. They are also used in many current biotechnologies such as biofuel production. The spatial distribution of elements, namely Cu, Ca, Ag, and silver nanoparticles, were measured on transverse sections of the plant shoots and roots using the LIBS double-pulse system in the orthogonal arrangement. The optimal method for the preparation of plant cross sections has been found allowing achieving a resolution of up to 50 μm and a sensitivity of LIBS measurements at the level of natural occurrence of these elements in the environment. The LIBS system in the double-pulse configuration was also used to rapid detection of metals in algal biofilms and algal suspensions. In the next part of this work the concentrations of elements in leaves and stems of polar willow (Salix polaris) were determined. The soils under the sampled plant of Salix polaris were analyzed for the same elements too. The samples were brought from the former Soviet mining town of Pyramiden and its close vicinity on the Arctic archipelago of Svalbard. The assessment of the contamination of soils and plants by toxic heavy metals in an otherwise pristine environment of the Svalbard archipelago related to urban/industrial activities were done.
