Analysia Optimalization of the Determination of Free Haemoglobin in Plasma

Abstract:

V této diplomové práci bylo testováno stanovení koncentrace volného hemoglobinu v plazmě metodou pomocí Drabkinova transformačního činidla na analyzátoru ELAN, metodou podle Harboea na analyzátoru ELAN a spektrofotometru SPEKOL 11. Vzájemně se metody liší použitím rozdílných vlnových délek pro měření absorbancí a koncentrací, dále použitými reagenciemi a tím i výslednou citlivostí stanovení koncentrací nalezeného volného hemoglobinu v plazmě. Byly zjištěny vyšší koncentrace volného hemoglobinu v plazmě stanovením pomocí Drabkinova transformačního činidla než metodou podle Harboea.

Abstract:

There was tested the determination of free haemoglobin concentrations in plasma by the metod of using the Drabkin?s transformation reagent on the ELAN analyser and by the metod of Harboe on the ELAN analyser and spectrophotometer SPEKOL 11. The methods differ in the wave lengths, in the used chemicals and also finaly by the sensitivity for free haemoglobin concentrations estimated in plasma. Higher concentrations of free haemoglobin in plasma were found by the use of the of Drabkin?s transformation reagent in relation to the metod of Harboe.
