Evaluation of the effect of the application of chelating agents on the ac-cumulation of heavy metals in Cannabis sativa L. cultivars.

Abstract:

V této diplomové práci popisuji schopnost rostlin dekontaminovat životní pro-středí od cizorodých látek. Primárně se zaměřuji na organické polutanty a těžké kovy. Popisuji metody, kterými rostliny tyto látky přijímají a odbourávají a fytoremediační technologie, kterými se dají jednotlivé skupiny látek odstraňovat ze životního prostředí. Literární rešerš obsahuje taky několik kapitol o látkách, které mají schopnost zlepšovat potenciál fytoextrakce kontaminantů a snižovat fytotoxický efekt kontaminujících látek na rostliny, které je přijímají. V další části této diplomové práce se zaměřuji na dva pokusy. První pokus zkoumá fytotoxicitu kyseliny citronové a kyseliny nitrilotrioctové při různých koncentracích na dvě odrůdy technického konopí (Cannabis sativa L.). Cílem tohoto pokusu bylo najít takovou aplikační dávku těchto látek, která nemá průkazně fyto-toxický efekt na rostliny, které mají dekontaminovat potenciální stanoviště. Druhým pokusem bylo zjišťování potenciálu fytoextrakce zinku z kontamino-vané půdy pomocí dvou registrovaných a jedné neregistrované odrůdy konopí (Cannabis sativa L.). V tomto pokus byla do půdy přidávaná kyselina citronová a kyselina nitrilotrioctová v koncentracích, které vyšli jako optimální v první pokusní části této práce pro podporu fytoextrakce. Výsledkem tohoto pokusu je zjištění, na kolik tyto látky dokáží zlepšit schopnost fytoextrakce zinku a porovnání fytotoxického efektu těchto látek a nadměrné koncentrace zinku v půdě. Výsledky ukázaly že aplikace NTA dokáže zvýšit koncentraci zinku v nad-zemních částech rostlin až o 300 % v porovnání s kontrolní skupinou. Neregistrovaná odrůda byla schopna do nadzemních částí absorbovat nejvíc zinku. Nejméně ze všech testovaných odrůd dokázala absorbovat odrůda Monoica. Tato odrůda dokázala u skupin ošetřených NTA absorbovat jen poloviční množství zinku.

Abstract:

In this diploma thesis I describe the ability of plants to decontaminate the environment from pollutants. This work primarily focuses on organic pollutants and heavy metals. I describe the methods, by which plants receive and degrade these substances and phytoremediation technologies, by which individual groups of substances can be removed from the environment. The literature review also contains several chapters on substances (amendments), that have the ability to improve the potential for phytoextraction of contaminants and reduce the phytotoxic effect of contaminants on the plants that receive them. In the next part of this diploma thesis I focus on two experiments. The first experiment investigates the phytotoxicity of citric acid and nitrilotriacetic acid at different concentrations on two varieties of industrial hemp (Cannabis sativa L.). The aim of this experiment was to find such an application dose of these substances that does not have a significant phytotoxic effect on plants. The second experiment was to determine the potential for phytoextraction of zinc from contaminated soil using two registered and one unregistered variety of cannabis (Cannabis sativa L.). In this experiment, citric acid and nitrilotriacetic acid were added to the soil in concentrations that turned out to be optimal in the first experimental part of this work to promote phytoextraction. The result of this experiment was to determine how much these substances can improve the ability of phytoextraction of zinc and to compare the phytotoxic effect of these substances and excessive concentrations of zinc in the soil. Results shown that NTA application can increase concentration of zinc in shoots by 300 %, compared to control group. Unregistered variety showed to be the most effective in absorption of zinc in shoots. Least successful variety in phytoextraction of zinc was Monoica, which was able to absorb just half of zinc content in group treated with NTA in comparison to other varieties.
