Ecophysiological aspects of eradication of mistletoe using herbicides

Abstract:

Viscum album L. je poloparazitická rostlina parazitující na mnoha různých druzích dřevin po celém světě. Jmelí hostiteli díky svým haustoriím bere značnou část vody a minerálních látek. V bakalářské práci byl zjišťován fyziologický mechanizmus působení dvou různých herbicidů na rostliny jmelí. První herbicid s názvem Ethrel, s účinnou látkou Ethephon, jejímž rozkladem se uvolňuje rostlinný hormon etylen, druhý herbicid s názvem Dicopur M750 s účinnou látkou kyselina 2,4-dichlorfenoxyoctová (2,4-D), patřící do skupiny rostlinných hormonů auxinů. Byly sledovány tyto ekofyziologické parametry – fluorescence chlorofylu, vodní potenciál a spektrální reflektance – v období od postřiku jmelí až do odumření rostlin. Měření fluorescence chlorofylu ukázalo, že u rostlin po aplikaci herbicidu s účinnou látkou Ethephon došlo k výraznému poklesu měřených hodnot indikující poškození světelné části fotosyntézy, na rozdíl od jmelí, na něž byl aplikován herbicid Dicopur M750. Při měření vodního potenciálu došlo u rostlin postříkaných herbicidem Ethrel k většímu poklesu hodnot vodního potenciálu a tím pádem i k většímu úbytku volné vody v rostlinných pletivech, než tomu bylo u rostlin postříkaných herbicidem Dicopur M750, kde byl sledován pouze mírný pokles hodnot vodního potenciálu. Rostliny tak v tomto případě netrpěly nedostatkem vody až do posledního týdne před úmrtím. Při měření spektrální reflektance bylo měřeno šest indexů, které nejlépe ukazovaly rozdíly při měření, konkrétně indexy MCARI, MCARI1, ARI2, PRI, RDVI a NDVI. U indexů MCARI, MCARI1 a RDVI došlo u obou herbicidů k podobnému poklesu hodnot, což indikuje působení herbicidů na světelnou část fotosyntézy v případě herbicidu Ethrel a působení na rozpad buněčných membrán rostliny díky herbicidu Dicopur M750. U indexů ARI2, PRI a NDVI byly sledovány rozdílné hodnoty u obou herbicidů – byl sledován rozdílný obsah cytokininů, antokyanů a obsah chlorofylu v listech jmelí. Výsledky ukazují, že díky oběma herbicidům došlo k celkovému úmrtí všech rostlin jmelí. Rozdíl je v mechanizmu působení herbicidů. Herbicid Ethephon pravděpodobně zapříčinil nadměrné otevření průduchů rostlin jmelí a následně pak došlo k vytranspirování veškeré vody a úmrtí. Herbicid 2,4-D pak s největší pravděpodobností zapříčinil rozpad buněčných membrán rostliny. Herbicid na bázi 2,4-D tak potencionálně může poškodit i buněčné membrány v haustoriích a omezit tak regeneraci jmelí z haustorií.

Abstract:

Viscum album L. is a hemi-parasitic plant, which parasites on many different tree species worldwide. Mistletoe takes from its host a considerable amount of water and nutrients by its haustoria. In the bachelor thesis, I investigated the physiological mode of herbicide action on mistletoe plants. The effects of two herbicides were studied. The first herbicide named Ethrel, with the active substance Ethephon, dissolving into a plant hormone ethylen. The second herbicid called Dicopur M750 with the active substance 2,4-dichlorophenoxyacetic acid (2,4-D), which is a synthetic plant hormone auxin. The following ecophysiological parameters – chlorophyll fluorescence, water potential and spectral reflectance were studied. Measurement of chlorophyll fluorescence showed its significant decrease shortly after the application of the Ethephon herbicide, which was not observed after the application of Dicopur M750. The decrease of the dark fluorescence indicated a permanent damage to the light part of the photosynthesis. Water potential decreased more in plants sprayed with Ethrel, than in the plants treated with Dicopur M750. Sharp increase in the water potential just before the death together with drop in the chlorophyll fluorescence indicates a damage to the cell membranes in trees treated with Dicopur M750. Indexes of spectral reflectance MCARI, MCARI1 and RDVI indexes indicated the action of the herbicide on the light part of the photosyntesis in the case of herbicide Ethrel and the action of the breakdown of the cell membranes of the plant by the herbicide Dicopur M750. ARI2, PRI and NDVI indexes indicated different effect of the two herbicides on cytokinins, anthocyanins and amount of chlorophyll in mistletoe leaves. Both herbicides caused total death of all mistletoe plants. The difference was in the mechanism of action of herbicides. Herbicide based on Ethephon caused opening of stomata and loss of water with subsequent death. Herbicide 2,4-D then most likely caused the disintegration of cell membranes. Because of herbicide based of 2,4-D damage cell membranes, which are in all plant organs, it has a greater potential to kill entire plant including haustoria, than Ethephon.
