Digestive tract microflora of stag beetle (Lucanus cervus L.)

Abstract:

Práce má rešeršní i experimentální charakter. Cílem rešeršní části je shrnout současné znalosti o rozkladu dřevní hmoty v přírodě v souvislosti s dostupnými informacemi o střevní mikroflóře dřevokazných druhů hmyzu a nastínit některé současné metody studia mikrobiálních společenstev. Cílem experimentální části je zajistit vzorky střevní mikroflóry larev roháče obecného (Lucanus cervus L.) a následně se pokusit z nich kultivovat a molekulárně identifikovat jejich zástupce. Dalším cílem bylo popsat prokaryotní, eukaryotní a houbová společenstva exkrementů larev pomocí masivně paralelního sekvenování na platformě Illumina. Bylo zjištěno, že mezi eukaryotními mikroorganismy exkrementů dominují kvasinky rodu Scheffersomyces, pravděpodobně náležející druhu Scheffersomyces stipitis. Byl získán izolát tohoto druhu kvasinky, který je udržován v kultuře. Mezi prokaryotními mikroorganismy dominovaly bakterie řádu Clostridiales.

Abstract:

This thesis has the character of literature review complemented by experimental section. The aim of the review part is to summarize the existing knowledge about wood decomposition in nature in connection with available information about the intestinal microflora of wood-boring insect species and also to tract some current methods of studying microbial communities. The aim of the experimental part is to obtain the samples of intestinal microflora of stag beetle (Lucanus cervus L.) larvae and to attempt the isolation and molecular characterization of representative isolates. The other aim is to describe prokaryotic, eukaryotic and fungal communities of the excrements using high throughput sequencing on the Illumina platform. Scheffersomyces yeasts, probably Scheffersomyces stipitis, dominated the eukaryotic component of the excrement microbial community. A strain of this yeast was isolated and is maintained in culture. The prokaryotic community was dominated by representatives of the bacterial order Clostridiales.
