Magnetoreception under long wavelength light.

Abstract:

Magnetorecepce zvířat a citlivost některých biochemických reakcí na magnetické pole Země není zatím objasněným fenoménem. Je známo, že citlivost k magnetickému poli Země je pravděpodobně postavena na reakcích aktivovaných světlem, kdy vznikají radikálové páry schopné s tímto polem interagovat. Otázka spektrální citlivosti magnetorecepce zvířat představuje důležitý klíč k pochopení otázky, jaké molekuly spolu interagují a jak je signál předáván. U některých živočišných druhů byla popsána magnetorecepce i za dlouhých vlnových délek, která je těžko vysvětlitelná pomocí klasické představy o fotoaktivaci kryptochromů. Předkládaná diplomová práce se zabývá vlivem různých intenzit a vlnových délek světla zejména z dlouhovlnné části spektra na magnetorecepci ruměnice pospolné. Jako zkoumaný organismus byl zvolen hmyz, ruměnice pospolná (Pyrrhocoris apterus), která je dobře prozkoumaným druhem využívajícím kryptochromovou signalizaci pro řízení cirkadiánních rytmů, o kterém však nejsou žádné poznatky týkající se magnetorecepce. První část práce je věnována rozpracování a optimalizaci nového behaviorálního testu, který je založen na měření aktivity jedinců v závislosti na periodické rotaci magnetického pole. Sledovali jsme magneticky vyvolaný freezing jako podmíněnou motorickou odpověď na magnetický stimul při různých světelných podmínkách. Použili jsme širokospektrou žárovku a monochromatické osvětlení s maximem při vlnových délkách 505, 526, 595 a 635 nm a odlišnými intenzitami. Protože magnetorecepční fenotyp se objevil pouze u samců, byly provedeny dílčí experimenty usilující o vysvětlení tohoto rozdílu. Výsledek práce je funkční test a doporučené parametry hodnocení freezingu ruměnic. Citlivost k magnetickému poli se objevila při žárovce a světle s vlnovou délkou 505 nm. Při vyšších vlnových délkách a intenzitách se ztratila s výjimkou 595 nm jedné intenzity. Při porovnání s dříve získanými výsledky naší laboratoře u rusa domácího (Blattella germanica) není odpověď na rotaci magnetického pole při stejných světelných podmínkách totožná a je možné, že tento rozdíl je právě způsoben odlišnou kryptochromovou výbavou.

Abstract:

Magnetoreception in animals and sensitivity of some biochemical reactions to the Earth´s magnetic field is a phenomenon that has not been clarified so far. It is known that the sensitivity to the EMF is probably based on the reactions activated by light when radical pairs able to interacted with the Earth´s magnetic field are formed. The issue of spectral sensitivity of magnetoreception of animals represents an important clue to understand the question as to which molecules interact with each other and how the signal is transferred. In some animal species, the magnetoreception has been described even under long-wavelength light, which is difficult to explain through the traditional notion of photoactivation of cryptochromes. The presented thesis deals with the influence of various intensities and wavelengths of light, particularly from the long-wavelength part of the spectrum, on the magnetoreception of the firebug. As an organism to examine, we have chosen the firebug (Pyrrhocoris apterus), which is a well-examined species that uses the CRY signaling to control circadian rhythms; however, no knowledge of its magnetoreception is available. The first part of the thesis is devoted to the preparation and optimisation of a new behavioural test that is based on the measurement of activity of individuals depending on the periodical rotation of the magnetic field. We have observed the magnetically evoked freezing as a conditioned motoric response to the magnetic stimulus under different light conditions. We have used a full-spectrum light and monochromatic light with its maximum at a wavelength of 505, 526, 595 and 635 nm and different intensities. Since the magnetoreception phenotype was only detected in males, partial experiments attempting to clarify this difference have been made. The thesis outcomes include a functional test and recommended parameters of the evaluation of freezing of firebugs. The sensitivity to the magnetic field has been detected under full-spectrum light and 505 mm light. Under long wavelengths and higher light intensities, the sensitivity has disappeared except for 595 nm light at one intensity. When compared to the results of our laboratory previously obtained in the German cockroach (Blattella germanica). The response to the rotation of the magnetic field under the same light conditions is not the same, and it is possible that this difference is just caused by a different cryptochrome composition.
