Abstract:

Předkládaná disertační práce byla zaměřena na studium cytoskeletu, ultrastruktury a morfologie dvou potenciálně patogenních, bazidiomycetových kvasinek – Malassezia pachydermatis a Cryptococcus laurentii. Z cytoskeletálních struktur byly imunofluorescenčně u obou mikroorganizmů identifikovány mikrotubuly a byla poznána jejich distribuce a dynamika v průběhu buněčného cyklu. V buňkách byly pozorovány cytoplazmatické mikrotubuly během interfáze, difúzní fluorescence v pupenu před započetím mitotického dělení a dělící vřeténko s astrálními mikrotubuly v M-fázi. U obou studovaných druhů bazidiomycetových kvasinek byl zjištěn přesun jádra do pupenu před jeho rozdělením. Pro vizualizaci mikrotubulů u Malassezia pachydermatis byla nezbytná prodloužená inkubace buněk se stěnovými lytickými enzymy. Fluorescenční a imunofluorescenční barvení aktinového cytoskeletu Malassezia pachydermatis se nezdařilo. Aktin byl u této kvasinky identifikován pouze imunoznačením na ultrastrukturální úrovni. Zjištěný aktin byl převážně asociován s plazmatickou membránou. U opouzdřeného kmene Cryptococcus laurentii byly imunofluorescenčním značením prokázány aktinové tečky, lokalizované zejména v rychle rostoucích částech buněk a během cytokineze při bázi pučení. Při fluorescenčním barvení rhodamin-faloidinem nebyly v buňkách tohoto kmene aktinové struktury prokázány. Barvivo se zřejmě vázalo na komponenty pouzdra. Fluorescenčně byl F-aktin identifikován a jeho distribuce a dynamika byly poznány v průběhu buněčného cyklu neopouzdřeného kmene Cryptococcus laurentii. Absence pouzdra na buněčném povrchu byla u tohoto kmene potvrzena jak na tušových preparátech, tak pomocí rutheniové červeně na ultratenkých řezech. V buňkách se během celého buněčného cyklu nacházely aktinové tečky a to především akumulované v místech intenzivního růstu. U pučících buněk byla identifikována aktinová vlákna, která z mateřské buňky směřovala nejprve do krátké stopky a následně do rostoucího pupenu, jenž se na stopce vytvářel. Po dokončení mitotického dělení aktinová vlákna zanikla a při bázi pučení se vytvořil cytokinetický prstenec. Rozložení a dynamika aktinového cytoskeletu Cryptococcus laurentii byly podobné jako u příbuzného druhu Cryptococcus neoformans. U Cryptococcus laurentii však byl F-aktin chudší. Stádia buněčného cyklu, která byla u studovaných druhů rozlišitelná prostřednictvím fluorescenční mikroskopie, byla detekována také na ultrastrukturální úrovni, včetně přesunu jádra do pupenu před zahájením mitotického dělení. Na rozdíl od Cryptococcus laurentii pučely buňky Malassezia pachydermatis monopolárně na široké bázi a nevytvářela se na nich stopka. Pupen se u tohoto druhu odděloval od mateřské buňky štěpením. Na povrchu buněk Malassezia pachydermatis nebyly vyjma límečku zaznamenány žádné jizvy po předchozích pučeních. U Cryptococcus laurentii byl límeček méně výrazný a na povrchu buněčné stěny se vyskytovaly další jizvy. V límečku buněk obou studovaných kvasinek a v jizvách u Cryptococcus laurentii byl pravděpodobně akumulován chitin, soudě podle intenzivní fluorescence v těchto místech při barvení buněčné stěny a afinitě použitých barviv k chitinu. Oproti Cryptococcus laurentii měly buňky Malassezia pachydermatis buněčnou stěnu silnou a kompaktní. Bylo potvrzeno, že buněčná stěna obou kvasinek má lamelární charakter a jsou v ní rozlišitelné jednotlivé vrstvy. Přítomnost partikulí ve vnější vrstvě buněčné stěny Cryptococcus laurentii by mohla souviset s produkcí extracelulárních polysacharidů. ěna. Septum buněk Cryptococcus laurentii bylo elektron-denzní méně. Jeho denzita byla srovnatelná s denzitou vnější vrstvy buněčné stěny této kvasinky. U Malassezia pachydermatis byla potvrzena jedinečná organizace vnitřního povrchu buněčné stěny a plazmatické membrány. Na vnitřní straně buněčné stěny se nacházely hřebeny, uspořádané do spirál, které korespondovaly se spirálovitě uspořádanými vchlípeninami plazmatické membrány. Absence těchto struktur byla zaznamenána při bázi pučení a na vrcholu pupenu. U buněk Cryptococcus laurentii se tato jedinečná organizace povrchů nevyskytovala. Byly zde pouze jednotlivé vchlípeniny (invaginace) plazmatické membrány, které byly rozmístěny nahodile. Další dvě specifické struktury, které byly na plazmatické membráně buněk Malassezia pachydermatis potvrzeny, byly ohrazené výdutě a prstencovité zduření. Ohrazené výdutě se nacházely při bázi pučení mezi sousedními vchlípeninami membrány. V místě jejich výskytu byla buněčná stěna kvasinky nejtenší. Předěl mezi mateřskou buňkou a pupenem u tohoto mikroorganizmu tvořilo prstencovité zduření plazmatické membrány. Buňky Cryptococcus laurentii ohrazené výdutě a prstencovité zduření plazmatické membrány postrádaly. V buňkách obou studovaných druhů byly potvrzeny organely typické pro kvasinky - jádro, mitochondrie, vakuoly, endoplazmatické retikulum a Golgiho aparát. Dále byly pozorovány ribosomy v cytoplazmě, jaderné póry v jaderné membráně a kristy uvnitř mitochondrií. U buněk Malassezia pachydermatis byla mimoto zjištěna akumulace malých měchýřků při vrcholu pupenu a přítomnost drobných veziklů mezi sousedními vchlípeninami plazmatické membrány. Jadérko bylo detekováno pouze u Cryptococcus laurentii. Distribuce mikrotubulů identifikovaná v průběhu buněčného cyklu obou studovaných druhů je obdobná jako u dalších bazidiomycetových kvasinek. Aktinový cytoskelet Cryptococcus laurentii obsahuje všechny tři typy F-aktinových struktur, které jsou pro kvasinky typické. V buňkách Malassezia pachydermatis by prokázaný aktin mohl souviset s jedinečným uspořádáním plazmatické membrány a vnitřního povrchu buněčné stěny. Potíže při vizualizaci jednotlivých složek cytoskeletu studovaných druhů jsou patrně zapříčiněny extracelulárními strukturami buněk. V případě Malassezia pachydermatis je zřejmě příčinou těchto potíží silná buněčná stěna a u Cryptococcus laurentii pouzdro.

Abstract:

In the present thesis, the microtubular and actin cytoskeletons, ultrastructural characteristics and morphology of two potentially pathogenic basidiomycetous yeasts Malassezia pachydermatis and Cryptococcus laurentii were investigated. At studied species, the distribution of microtubules was described in relation to nucleus position and bud development. By indirect immunofluorescence, cytoplasmic microtubules were detected in interphase and a spindle with astral microtubules was seen in M-phase. The disintegration of cytoplasmic microtubules and migration of the nucleus to the bud before mitosis were characteristic features of both basidiomycetous yeasts. To detect microtubules in Malassezia pachydermatis cells, a prolonged incubation with lysing enzymes was necessary. The visualisation of F-actin structures in Malassezia pachydermatis cells by fluorescence microscopy, using both monoclonal anti-actin antibody and rhodamine-phalloidin, failed. However, actin was detected by electron microscopy with immunogold labelling. Clusters of gold particles indicating actin structures were detected mainly at the plasma membrane. A possible association of the actin cytoskeleton with unique cortical features characteristic of the genus Malassezia was suggested. The detection of actin cytoskeleton in an encapsulated strain of Cryptococcus laurentii by rhodamine-phalloidin was not successful. A possible explanation is a potential non-specific interaction of rhodamine-phalloidin with capsular polysaccharides. Using indirect immunofluorescence, actin patches were demonstrated in encapsulated strain cells. During budding, they were accumulated in growing bud and after nuclear division, they occurred at the budding base. The distribution of F-actin structures during the cell cycle of an acapsular strain Cryptococcus laurentii was described. The absence of a capsule was confirmed by India ink staining and by ruthenium red staining in ultrathin sections. Using rhodamine-phalloidin, all structures typical of yeast cells were observed, i.e., actin patches detected in the course of whole cell cycle, actin cables extending from the mother cell to sterigma and subsequently to growing bud and cytokinetic ring during cytokinesis. The distribution of actin cytoskeleton in Cryptococcus laurentii was similar to those in Cryptococcus neoformans, but poorer. All stages of cell cycle, as detected by fluorescence microscopy, were also seen in both species at the ultrastructural level, including migration of the nucleus to the bud before mitosis. In contrast to Cryptococcus laurentii cells, the budding of Malassezia pachydermatis was monopolar on a broad base without development of sterigma and the bud separated from the mother cell by fission. On surface of Malassezia pachydermatis cells, apart from collar at the budding base, no bud scars were observed. Collar of Cryptococcus laurentii cells was not so marked. At this yeast, other bud scars were also presented on the cell wall surface. In both, the collar and the bud scars, chitin was probably accumulated there, inferring due to a strong fluorescence at these sites after staining of the cell wall and affinity of used dyes to chitin. In comparison with Cryptococcus laurentii, the Malassezia pachydermatis cells showed relatively thick and compact cell wall. The cell wall of both yeasts was multilamellar with some discernible layers. The presence of particles in the rough outer layer of Cryptococcus laurentii cell wall could be probably associated with production of extracellular polysaccharides. At Malassezia pachydermatis cells, the density of centripetal growing septum that was forming during cytokinesis was similar as the density of the cell wall. The septum of Cryptococcus laurentii cells was more electron-transparent. Its density was comparable to the density of outer cell wall layer. e unique pattern. Right-handed spiral ridges on the inner side of the cell wall corresponded to left-handed spiral grooves of the plasma membrane. They were missing only on the broad budding base and on the bud apex. In Cryptococcus laurentii cells, the unique pattern of spiral grooves was absent and the plasma membrane invaginations were distributed irregularly. Two further unusual structures were confirmed on the plasma membrane of Malassezia pachydermatis cells. Near the budding base between neighbouring grooves of the plasma membrane, circumvallate bulgings were detected. The cell wall in these regions was the thinnest. Moreover, the plasma membrane of this yeast formed a ring-like swelling in the budding base (between the mother cell and the bud). Both, the circumvallate bulgings and the ring-like swelling, were absent on the plasma membrane of Cryptococcus laurentii cells. The presence of organelles typical for yeasts, i.e., nucleus, mitochondria, vacuoles, endoplasmic reticulum and Golgi apparatus, in cells of investigated species was confirmed. Furthermore, ribosomes in cytoplasm, nuclear pores in nuclear envelope and cristae inside mitochondria were discernible. In Malassezia pachydermatis cells, small vesicles were detected at the apex of the growing bud and between neighbouring grooves of plasma membrane. The nucleolus was revealed only at Cryptococcus laurentii. At studied species, the distribution and the dynamics of identified microtubules are similar to those in other basidiomycetous yeasts. In Cryptococcus laurentii cells, all F-actin structures characteristic of yeast cells were observed, but the actin equipment is poorer than that one of Cryptococcus neoformans. In Malassezia pachydermatis cells the demonstrated actin may be associated with unique pattern of the plasma membrane and corresponding inner cell wall surface. The failure of cytoskeletal structures’ detection in studied yeasts can be related to their extracellular structures. At Malassezia pachydermatis, the most likely cause is the thick cell wall and at Cryptococcus laurentii it is the capsule.
