Use of continuous flow bioreactor for control of homeostasis of the intracellular environment in the course of in-cell NMR experiment.

Abstract:

Pro studium struktur proteinů a nukleových kyselin se v posledních letech využívá metoda in-cell NMR, která umožňuje tyto látky zkoumat v jejich přirozeném prostředí. Absence regulovaných podmínek v dlouhotrvajícím in-cell NMR experimentu nepříznivě ovlivňuje stav buněk a limituje dobu měření. V této práci jsem testovala různé způsoby výměny média v NMR vzorku a různé nosiče buněk linie HeLa, které by umožňovaly rovnoměrný přístup média k buňkám. Metabolický stav buněk byl hodnocen na základě změn vnitrobuněčné hladiny ATP a pH pomocí NMR spektroskopie, viabilita buněk pak barvením trypan blue. Zalití buněk do spirál agarózového gelu SeaPrep a přivedení čerstvého média na dno NMR zkumavky, tak, aby protékalo celým vzorkem, zajistí stabilitu pH ve vzorku a zabrání úbytku ATP v buňkách po dobu nejméně 10 hodin.

Abstract:

In the past few years, the in-cell NMR method has been used to study the structures of proteins and nucleic acids inside the cells, i.e., in their natural environment. The absence of regulated culture conditions within a long-term in-cell NMR experiment adversely affects the condition of the cells and limits the measurement time. In this work, I tested different methods of medium exchange in the NMR sample and different carriers of the HeLa cells, which would allow uniform access of medium to the cells. The metabolic state of the cells was evaluated based on changes in the intracellular ATP level and pH by NMR spectroscopy, and the viability of the cells by trypan blue staining. Embedding the cells into SeaPrep agarose gel threads and bringing the fresh medium to the bottom of the NMR tube so that it flows through the entire sample will ensure pH stability in the sample and prevent ATP loss in the cells for at least 10 hours.
