Cloning of lectins from Photorhabdus spp.

Abstract:

Lectins are proteins of a nonimmune origin able to bind to sacharides specifically and reversibly. They occur throughout the whole spectrum of living systems and serve different functions. While the animal lectins are an important part of the immune system and help to roll leukocytes towards the site of the inflammation, the bacterial lectins, often expressed on the surface of microorganisms function as adhesins that bind to host's surface glycans. The adhesion of bacteria leads to an infection. Anti-adhesive therapy is one of the possible strategies in the prevention and treatment of bacterial infections. In this thesis we deal with hypothetical lectins from the Photorhabdus luminescens bacterium that lives in a symbiotic relationship with nematodes from the Heterorhabditis genus. The nematodes live in the soil where they search activelly for an insect victim. After penetrating the body of larva the symbiont is released and produces toxins that kill the insect. At our department a new lectin from Photorhabdus luminescens was discovered. According to bioinformatics analysis four hypothetical lectins were found from the bacterium with a sequence homology to the lectin. The thesis focuses on the preparation of constructs for expression of recombinant hypothetical lectins using molecular-biology methods. The thesis involves also a short analysis of hypothetical lectins using bioinformatics tools.

Abstract:

Lektíny sú proteíny neimunitného pôvodu, ktoré dokážu špecificky a reverzibilne viazať sacharidy. Vyskytujú sa naprieč celým spektrom živých organizmov, kde plnia rôzne funkcie. Zatiaľ čo živočíšne lektíny sú dôležitou súčasťou imunitného systému a pomáhajú nasmerovať leukocyty do miesta zápalu a infekcie, bakteriálne lektíny exprimované na povrchu mikroorganizmov sú súčasťou adhezívnych reakcií, ktoré vedu k napadnutiu a infikovaniu hostiteľa. Antiadhezívna terapia je jednou z možných stratégií pre prevenciu a liečbu bakteriálnych infekcií. V práci sa venujeme hypotetickým lektínom z baktérie Photorhabdus luminescens, ktorá žije v symbiotickom vzťahu s hlístovcami rodu Heterorhabditis. Hlístovce žijú v pôde, kde aktívne vyhľadávajú hmyziu obeť, ktorú s pomocou symbionta usmrtia. Na našom pracovisku bol objavený nový lektín z Photorhabdus luminescens. S využitím bioinformatických nástrojov boli nájdené štyri hypotetické lektíny, ktoré vykazujú pomerne vysokú sekvenčnú homológiu s uvedeným lektínom. Práca sa zaoberá prípravou konštruktov pre expresiu hypotetických lektínov v rekombinantnej forme s využitím molekulárno-biologických postupov. Súčasťou práce je aj analýza hypotetických lektínov s využitím bioinformatických nástrojov.
