Mutations in APP and PSEN2 genes in patients with early onset Alzheimer's disease

Abstract:

Alzheimerova choroba (AD) patří do skupiny neurodegenerativních onemocnění centrálního nervového systému a postihuje okolo 5 % osob nad 65 let. Rozlišujeme dvě formy Alzheimerovy choroby ? časnou a pozdní (s věkovou hranicí 65 let). Časná forma Alzheimerovy choroby souvisí s mutacemi v PSEN1, PSEN2 a APP genech. Skríning mutací v těchto genech je prováděn molekulárními metodami jako jsou PCR, elektroforéza a sekvenování. Práce obsahuje výsledky sekvenování níže uvedených genů u skupiny 9 pacientů s klinicky časnou formou AD. Průměrný věk pacientů je 57 let (44 - 69 let). Na genetické došetření byli pacienti posláni neurologem. DNA pacientů byla již dříve testována na přítomnost mutací v genu PSEN1 a ?hotspot? exonu 17 genu APP. Cílem této bakalářské práce bylo rozšířit genetické testování o skríning mutací v genu PSEN2 a v exonu 16 genu APP.

Abstract:

Alzheimer's disease (AD) belongs to a group of neurodegenerative disorders of the central nervous system and affects about 5% of people aged over 65. There are two forms of Alzheimer's disease ? early-onset and late-onset (before age 65 years). Early-onset Alzheimer's disease is linked to the mutations in the PSEN1, PSEN2 and APP genes. Mutation?s screening in this genes is investigated by molecular methods such as PCR, electrophoresis and sequencing analysis. This work contains the results from the sequencing analysis the genes mentioned above in the group of the 9 patients with early-onset AD. The average age of the patients is 57 years (44 - 69 years). For the genetic testing were patients sent by the neurologist. DNA of the patients was previously tested for the presence of mutations in the gene PSEN1 and ?hotspot? exon 17 of the APP gene. The aim of this work was to extend the genetic testing for mutation?s screening of the PSEN2 gene and exon 16 of the APP gene.
