New mechanisms of endocrine disruption - disruption of thyroid and glucocorticoid system

Abstract:

Cílem této bakalářské práce je shrnout poznatky o endokrinních disruptorech narušujících thyroidní a glukokortikoidní systém. V poslední době se stále více studií zabývá endokrinní disrupcí – porucha hormonálního řízení organismu vyvolaná exogenními faktory. Předmětem studií bývá často narušení androgenního a estrogenního systému, ale v současnosti se předmětem výzkumů stává i thyroidní systém. Studií o narušení glukokortikoidního systému zatím není mnoho. Práce je rozdělená na dvě části – teoretickou a praktickou. Teoretická část popisuje zatím známé mechanizmy narušení thyroidního a glukokortikoidního systému environmentálními chemikáliemi. Praktická část se zabývá aktivací a inhibicí drah glukokortikoidního (GR) a androgenního receptoru (AR) v buněčné linii MDA-kb2. Cílem praktické části bylo navrhnout experiment, který nám řekne, zda testovaná látka aktivuje GR nebo AR. K rozlišení byla použita látka flutamid, která inhibuje aktivitu AR, ale na GR by neměla mít žádný vliv. Při testování však došlo k problému s vhodnou koncentrací flutamidu, doporučovaná koncentrace vykazovala inhibici aktivace AR pouze v nižších koncentracích aktivátoru AR – dihydrotestosteronu (0,1 až 1 nM) a při použití vyšší koncentrace flutamidu došlo k neočekávané inhibici i látek aktivujících GR.

Abstract:

The aim of this bachelor thesis is to summarize the findings about endocrine disruptors of thyroid and glucocorticoid system. An increasing number of studies have been lately dealing with endocrine disruption - hormonal regulation disorder of the body caused by exogenous factors. The object of studies is often disruption of androgen and estrogen systems yet at present even thyroid system becomes the subject of research whereas studies on the disruption of the glucocorticoid system are not many. The thesis is divided into two parts - theoretical and practical. The theoretical part describes currently known mechanisms of glucocorticoid and thyroid disruption by environmental chemicals. The practical part deals with the activation and inhibition of glucocorticoid (GR) and androgen receptor (AR) signaling pathways in the cell line MDA-kb2. Goal of practical part was to design an experiment that tells us whether the test substance activates the GR or AR. For the distinction flutamide was used. It inhibits the activity of the AR but it should not have any effect on the GR. When tested, there was a problem with the appropriate concentrations of flutamide. The recommended concentration showed inhibition of AR’s activation only in lower concentrations of AR activator – dihydrotestosterone (0.1 to 1 nM) and when higher concentration of flutamide was used, there also occured an unexpected inhibition of substances activating GR.
