The effects of a short- and long-term (chronic) low-temperature stress on anatomic and photosynthetic parameters in model lichen and algal species from polar regions

Abstract:

The main focus of my diploma thesis is study of influence of low and freezing temperatures on the anatomical structure of two model species of Antarctic lichens Usnea antarctica and Usnea aurantiaco-atra, which was observed on cross-sections of thallus and evaluated as changes in the width of individual anatomical layers. The theoretical part provides an overview of the knowledge gained so far about the impact of cold and freezing temperatures on model organisms, lichens and algae, and about the mechanisms of their resistance. Main experiment focused on the anatomical changes is supplemented by two other experiments that investigate the resistance of the algae culture Trebouxia erici, which was isolated from the Antarctic lichen Usnea Antarctica, against the effects of low and subzero temperature. The experimentally obtained results show that the shock freezing of lichen thallus in a hydrated, i.e. physiologically active state, causes significant changes in radially arranged layers of the thallus. In particular, the upper cortical layer and the central part of the thallus respond most remarkably. Based on additional experiments, I concluded that the symbiotic alga Trebouxia erici has a temperature optimum for growth at the temperature approximately 15°C. Model alga is an extremophilic organism that can be physiologically active to the temperature as low as -25°C, at which its photosynthesis is still measurable by the methods based on in vivo measurements of chlorophyll fluorescence.

Abstract:

Hlavním zaměřením mé diplomové práce je studium vlivu nízkých a mrazových teplot na anatomickou stavbu dvou modelových druhů antarktických lišejníků Usnea antarctica a Usnea aurantiaco-atra, který byl sledován na příčných řezech stélkou a vyhodnocován jako změny v šířce jednotlivých anatomických vrstev. Teoretická část diplomové práce přináší přehled dosavadních poznatků o vlivu chladu a mrazu na modelové organismy lišejníků a řas a o mechanismech jejich rezistence. Hlavní experiment zaměřený na anatomické změny je doplněn o další dva experimenty, které zkoumají odolnost řasové kultury Trebouxia erici, která byla vyizolovaná z antarktického lišejníku Usnea antarctica, vůči působení nízkých a mrazových teplot. Z experimentálně získaných výsledků plyne, že šokové zmrazení stélek lišejníků v hydratovaném, fyziologicky aktivním stavu, způsobuje významné změny v radiálně uspořádaných vrstvách stélky, přičemž nejvýrazněji reagují zejména vrchní korová vrstva a centrální část stélky. Na základě doplňujících experimentů jsem dospěla k závěru, že symbiotická řasa Trebouxia erici má teplotní optimum pro růst v teplotě přibližně 15°C. Modelová řasa je přitom extremofilním organismem a dokáže být fyziologicky aktivní až do teploty -25°C, ve které je ještě její fotosyntéza měřitelná pomocí metod založených na in vivo měření fluorescence chlorofylu.
