Characterisation of sequence variants of 45S rDNA spacers in Arabidopsis thaliana

Abstract:

V genomu Arabidopsis thaliana existuje několik stovek kopií genů 45S rRNA, které jsou sekvenčně téměř identické. Každé dvě kopie jsou odděleny mezerníkem, nekódující sekvencí, ve které je naopak velká variabilita. Buňka vyrovnává dávku genů rRNA tím, že některé kopie umlčuje epigenetickými mechanizmy, které působí v oblasti mezerníků. Dosud není známo, co rozhoduje o tom, které kopie budou umlčeny. V regulaci kopií by mohla hrát roli sekvenční variabilita mezerníků. V této diplomové práci se věnuji charakterizaci sekvenčních variant mezerníků v rostlinách wild-type a v rostlinách mutantních v genu fas1, který kóduje podjednotku chaperonu histonů CAF1. Mutace fas1 vede k postupné ztrátě 45S rDNA během několika generací (Mozgová et al., 2010). Výsledkem této práce je unikátní soubor sekvencí mezerníků, na kterém byla detailně zmapovaná jejich variabilita.

Abstract:

In Arabidopsis thaliana genome there is several hundred of 45S rRNA gene copies. The gene copies have high sequential identity and they are separated from each other by intergenic spacer (IGS). IGS is noncoding sequence with high sequential variability. The cell regulates rRNA gene dosage by silencing some of the gene copies by epigenetic mechanisms which act upon IGS. It is still unclear what dictates which gene copy is silenced. IGS sequential variability could play a role in the rRNA gene silencing. In this thesis, I focus on characterisation of sequence variants of IGS in wild-type plants and fas1 mutants. fas1 codes one of the subunit of the CAF1 histone chaperone. fas1 mutation results in gradual loss of the 45S rRNA genes over several plant generations (Mozgová et al., 2010). This thesis presents a unique set of IGS sequences, which was analyzed in order to map sequence variability of IGS.
