Study of posttranslational modifications ABD domain of plectin

Abstract:

Tato bakalářská práce se zabývá studiem aktin-vazebné domény myšího plektinu. Teoretická část zahrnuje informace z odborné literatury, které jsou zaměřeny na popis aktin-vazebné domény a na studium interakcí domény s různými vazebnými partnery. Aktin-vazebná doména je cílem i několika post-translačních modifikací, čímž dochází k ovlivňování vlastností a struktury domény. I tato problematika je projednávána v teoretické části, která je zaměřena především na nitrosylaci, vazbu nízkomolekulárního ligandu fosfatidylinositol-4,5-bisfosfátu (PIP2) a proteolytické štěpení. Praktická část je věnována přípravě třech izoforem aktin-vazebné domény pro strukturní analýzu NMR. Zahrnuje optimalizaci exprese a purifikace proteinu a převedení analytu do roztoku, ve kterém splňuje podmínky pro provedení NMR strukturní analýzy. Optimalizace přípravy proteinu jako vzorku pro analýzu NMR je prvním krokem pro další práci věnující se sledování interakcí a modifikací právě metodou NMR. Naměřená dvoudimenzionální spektra HSQC tvoří podklad pro další třídimenzionální měření. Zjištěné údaje tak mohou přispět k objasnění vlivu post-translačních modifikací na strukturu a na vazebné schopnosti domény.

Abstract:

This bachelor thesis deals with study of the actin-binding domain of mouse plectin. The theoretical part includes information from literature, which are focused on describing the actin-binding domain and the study of the interaction domains with different binding partners. Actin-binding domain is also the aim of several post-translational modifications, thereby influencing the properties and structure of the domain. Even this issue is discussed in the theoretical part, which focuses primarily on the nitrosylation, binding of low molecular ligand phosphatidylinositol-4,5-bisphosphate (PIP2) and proteolytic cleavage. The practical part is devoted to the preparation of three isoforms of actin-binding domain to NMR structural analysis. It includes optimizing protein expression and purification, and conversion of an analyte in a solution, which complies with the conditions for NMR structural analysis. Optimization of protein as a sample for NMR analysis is the first step for further work devoted to monitoring the interaction and modification just by NMR. Measurement of two-dimensional HSQC spectrum forms the basis for the further three-dimensional measurement. Collected data may help to explain the impact of post translational modifications to the structure and binding properties of the domain.
