Genetic structure of Pedicularis palustris L.

Abstract:

Pedicularis palustris citlivě reaguje na změny rašeliništních ekotypů, které pro něj představují důležitá refugia. Se zhoršujícím se stavem rašelinišť ve střední Evropě se Pedicularis palustris stal kriticky ohroženým druhem, k jehož ochraně můžeme přispět například i detailnější molekulární studií druhu. Ve své bakalářské práci zkoumám genetickou diverzitu čtyř populací duhu Pedicularis palustris z oblasti střední Evropy. První část práce tvoří rešerše na téma sekvenování nové generace se zaměřením na sekvenační protokol RADseq a sekvenační platformu Illumina, které dále využívám pro účely svého výzkumu. V další části prezentuji jednotlivé kroky izolace DNA, přípravy knihovny a výsledky populačně genetických analýz. Výstupem mojí práce je zjištění, že čtyři populace druhu Pedicularis palustris vykazují různou míru genetické diverzity v závislosti na geografické vzdálenosti mezi populacemi. Fakt, že mezi populacemi neprobíhá genetický tok, je důležitým poznatkem ohledně šíření tohoto druhu, který může přispět při ochraně jeho recentních populací.

Abstract:

Pedicularis palustris is sensitive to changes in peat ecotypes, which are important refuges for it. With the deteriorating state of peatlands in Central Europe, Pedicularis palustris has become a critically endangered species. We can contribute to its protection, for example, by a more detailed molecular research of the species. In my bachelor's thesis I study the genetic diversity of four populations of Pedicularis palustris from Central Europe. First, I have done a research on the topic of next generation sequencing with a focus on the RADseq sequencing protocol and the Illumina sequencing platform, which I both used for the purpose of my thesis. In the following section, I describe the consecutive steps of DNA isolation and library preparation, while presenting the results of each individual step in the next chapter. I also comment on the results of relevant population genetic analyzes. The output of my work is the finding that four populations of Pedicularis palustris show different degrees of genetic diversity depending on the geographical distance between populations. The fact that there is no gene flow between populations is an important finding about the spread of this species, which can contribute to the preservation of its recent populations.
