Study of lectins from opportunistic human pathogens

Abstract:

Patogenní lektiny představují podstatnou část bakteriálních a plísňových infekčních mechanismů. Tato diplomová práce byla zaměřena nejprve na mutantní formu lektinu AFL1, který je považován za modelový systém pro studium komplexních interakcí. Hlavním zájmem bylo zlepšení jeho rozpustnosti pomocí různých molekulárně-biologických a biochemických metod. Určení specifity potenciálního lektinu BC2L-B-nt bylo druhým hlavním cílem. Příprava na glycan array analýzu vyžadovala místně-specifické značení tohoto proteinu pomocí BirA biotin ligasy. Ačkoli struktura celého lektinu BC2L-B zůstává neznámá, struktura Cterminální domény s vázáným ligandem byla vyřešena a je v této chvíli v procesu vylepšování. Výsledky této práce mohou pomoct k dosáhnutí celkové charakterizace mechanismů adhese a infekce, které využívají lektiny, a následně mohou pomoct k modelování a syntéze nových antiadhesivních léků.

Abstract:

Pathogenic lectins represent a substantial part of bacterial and fungal infection mechanisms. This diploma thesis has been focused firstly on a mutant form of AFL1 lectin, which is considered as a model system for the study of complex interactions. The main concern was an enhancement of its solubility using various molecular biology and biochemistry methods. Specificity determination of potential lectin BC2L-B-nt was the second main goal. Preparation for glycan array analysis required site-specific labelling of this protein with the BirA biotin ligase. Although the structure of the whole BC2L-B lectin remains unknown, the structure of the C-terminal domain with bound ligand was solved and at this point is in the refinement process. Results of this thesis can help to achieve a comprehensive characterisation of the lectin-exploiting adhesion and infection mechanisms and, subsequently, to model and synthesise novel antiadhesive drugs.
