The effect of colloidal silver on selected mechanisms of nonspecific immunity.

Abstract:

Nanočástice jsou částice o velikosti 1-100 nm. V posledních letech jsou stále častěji využívány pro své výjimečné fyzikální, chemické a biologické vlastnosti. Nanočástice stříbra mají vynikající antibakteriální vlastnosti. Nanočástice zlata mohou být v budoucnu použity pro cílený transport léčiv. O tom jaký vliv mají nanočástice na buňky lidského organismu se zatím ale mnoho neví. Tato práce se snaží objasnit vliv nanočástic na vybrané mechanismy nespecifické imunity. Konkrétně jsem zjišťovali vliv nanočástic stříbra a zlata na míru oxidativního vzplanutí neutrofilů v plné krvi, na baktericidní působení čistého lysozymu a hemolymfy a na morfologii krevních buněk. Výsledky našich pokusů ukazují, že nanočástice stříbra ve vysokých koncentracích (25 μg/ml reakční směsi) snižují chemiluminiscenční (CL) signál aktivované CL. Naopak při spontánní CL došlo ke zvýšení CL signálu, což může souviset se zvyšující se tvorbou hydroxylových radikálů potvrzenou pomocí chemických systému pro tvorbu ROS. V těchto systémech ovšem stříbro zároveň snižovalo tvorbu superoxidového radikálu. Nanostříbro (5 μg/ml reakční směsi; 25 μg/ml reakční směsi) také snižovalo baktericidní aktivitu čistého lysozymu i hemolymfy. Nanočástice zlata (s nejvyšší použitou koncentrací 15 μg/ml reakční směsi) nemělo v našich pokusech vliv ani na spontánní ani na aktivovanou CL. Zároveň také nedocházelo k ovlivnění baktericidní aktivity čistého lysozymu ani hemolymfy. Po působení nanostříbra i nanozlata byly pozorovány vakuolizace v buňkách neutrofilů.

Abstract:

Nanoparticles are particles of the size 1-100 nm. In recent years, they are increasingly used for their unique physical, chemical and biological qualities. The nanoparticles of silver have excellent antibacterial qualities. The nanoparticles of gold can be used for a targeted drug transportation in the future. But it is not yet very much known about the influence of nanoparticles on cells of human organisms. The aim of this work is to explain the influence of nanoparticles on selected mechanisms of non-specific immunity. Specifically, it was being found out the effect of silver and gold nanoparticles on oxidative burst of neutrophils in whole blood, on bactericidal activity of pure lysozyme and hemolymph and on morphology of blood cells. The results of our experiments demonstrate that the silver nanoparticles at high concentration (25 μg/ml reactive mixture.) reduce chemiluminescent (CL) signal of activated CL. On the other hand, during spontaneous CL the CL signal was increased which may be related with the increasing production of hydroxyl radicals which is proved by a chemical system for the production of ROS. But in these systems silver reduced the superoxid radical production, as well. Nanosilver (5 μg/ml reactive mixture; 25 μg/ml reactive mixture) also decreased bactericidal activity of pure lysozyme and hymolymph. In our experiments, the gold nanoparticles (with the highest concentration of 15 μg/ml reactive mixture) did not have any influence on spontaneous nor activated CL. Bactericidal activity of pure lysozyme and hemolymph was not influenced, either. After the nanosilver and nanogold impact vacuolation in neutrophil cells were observed.
