Advanced 2D in vitro model as a tool for studies of pro-inflammatory effects of lipopolysaccharides of cyanobacterial water blooms

Abstract:

Cyanobacterial water bloom is a source of many secondary metabolites with negative effects on organisms, which come into contact with it. At the same, it is also a source of endotoxin, lipopolysaccharide. This endotoxin is a part of a cell wall of cyanobacteria and Gram-negative bacteria, microorganisms, which typically form the water bloom. It has been observed that cyanobacterial lipopolysaccharide can invoke eyes and skin irritation, allergies, headache, nausea, or respiratory and gastrointestinal diseases in humans. This thesis presents a study of the effects of the lipopolysaccharides isolated from cyanobacterial water bloom. Specifically it is aimed to monitor the proinflammatory effects of the lipopolysaccharides isolated from the water blooms found in the Czech Republic. To monitor these effects, an advanced 2D in vitro model of the intestinal epithelium was used, optimization of which has also been one of the aims of the thesis. The employed model represents coculture of intestinal epithelial cells Caco-2 and primary mononuclear cells isolated from peripheral blood. Utilizing this model, results revealing the level of activity of proinflammatory mechanisms induced by effects of some of the applicated cyanobacterial water bloom lipopolysaccharides were obtained. A part of the thesis is also an literature overview presenting 2D in vitro experimental models, intestinal epithelia and some manifestations of its inflammation, immune system of the intestine, cyanobacterial water bloom and a molecule of lipopolysaccharide.

Abstract:

Sinicový vodný kvet je zdrojom množstva toxických sekundárnych metabolitov s negatívnymi účinkami na organizmy, ktoré s ním prídu do styku. Zároveň je tiež zdrojom endotoxínu lipopolysacharidu. Tento endotoxín je súčasťou bunkovej steny siníc a Gram-negatívnych baktérií, mikroorganizmov typicky tvoriacich vodný kvet. Bolo zaznamenané, že sinicový lipopolysacharid je u ľudí schopný vyvolať podráždenia očí a kože, alergie, bolesti hlavy, nevoľnosť, či ochorenia respiračného a tráviaceho traktu. Táto práca prezentuje štúdium efektov lipopolysacharidov izolovaných zo sinicových vodných kvetov. Konkrétne je zameraná na sledovanie prozápalových efektov lipopolysacharidov izolovaných z vodných kvetov nájdených v Českej republike. Na sledovanie týchto efektov bol využitý pokročilý 2D in vitro model črevného epitelu, ktorého optimalizácia bola taktiež jedným z cieľov tejto práce. Použitý model predstavuje kokultúru črevných epitelových buniek Caco-2 a primárnych mononukleárnych buniek izolovaných z periférnej krvi. S využitím tohto modelu boli získané výsledky prezentujúce stupeň aktivity prozápalových mechanizmov vyvolaných efektom niektorých z aplikovaných lipopolysacharidov sinicových vodných kvetov. Súčasťou práce je aj literárna rešerš zaoberajúca sa 2D in vitro experimentálnymi modelmi, črevným epitelom a niektorými prejavmi jeho zápalu, imunitným systémom čreva aj sinicovým vodným kvetom a molekulou lipopolysacharidu.
