Stability complement system of hereditary angioedema

Abstract:

Komplementový systém tvoří jednu z hlavních součástí vrozené imunity a současně se podílí na vývoji adaptivní imunity. Onemocnění hereditární angioedém (HAE) vzniká deficiencí jedné z regulačních složek komplementového systému, respektive deficitem C1 inhibitoru. Hereditární angioedém je poměrně vzácné dědičné onemocnění, kterým v ČR trpí v současné době přibližně 200 osob. Cílem této práce je hodnocení stability komplementového systému ve vyšetřovaném séru u pacientů s hereditárním angioedémem a skupiny kontrolních osob v závislosti na čase od rozmrazení séra a také v závislosti na koncentracích některých složek komplementu. Stabilita komplementového systému je sledována pomocí testu CH50, tedy aktivace komplementu klasickou cestou a pomocí testu AH50, tedy sledování komplementového systému aktivovaného alternativní cestou.

Abstract:

The complement system is a major part of the innate immunity and also contributes to the development of adaptive immunity. Disease hereditary angioedema (HAE) arises from a deficiency of regulatory components of the complement system, more precisely C1 inhibitor deficiency. Hereditary angioedema is a relatively rare hereditary disease which currently suffering approximately 200 people in the Czech Republic. The aim of this thesis is to assess the stability of the complement system in the tested serum from patients with hereditary angioedema and group of control persons, depending on the time from thawing serum and also depending on the concentrations of some components of the complement. The stability of the complement system is monitored by a CH50 test, so activation of the complement by classical pathway and using test AH50, so monitoring of the complement system activated by alternative pathway.
