Study Toxicity Of Heavy Metals In Suspension Cell Systems

Abstract:

Předložená diplomová práce se zabývá studiem toxicity těžkých kovů in vitro pomocí pokročilých moderních bioanalytických metod. Teoretická část diplomové práce je zaměřena na působení těžkých kovů rtuti a kadmia. Dále jsou také zmíněny nejčastěji používané buněčné modely suspenzních buněk pro studium cytotoxicity těžkých kovů. Závěrečná pasáž teoretické části diplomové práce je zaměřena na využití moderních bioanalytických metod pro stanovení cytotoxicity těžkých kovů na lidských suspenzních buněčných liniích. Experimentální část práce shrnuje přípravu a kultivaci buněčných linií HL-60, Jurkat a Raji. Dále je diplomová práce věnována popisu cytotoxického působení CdCl2, HgCl2 a cisplatiny u zvolené buněčné linie Jurkat. K detekci funkčních změn buněk Jurkat inkubovaných s výše zmíněnými toxiny bylo využito stanovení koncentrace intracelulárního glutathionu a dehydrogenázové aktivity. Získané výsledky byly vyhodnoceny, diskutovány a následně v závěru diplomové práce shrnuty.

Abstract:

This diploma thesis deals with the study of heavy metal toxicity in vitro using advanced modern bioanalytical methods. The theoretical part of the thesis is focused on the effects of heavy metals such as mercury and cadmium. Furthermore, the most commonly used cell suspension models for the study of heavy metal cytotoxicity are also mentioned. The following theoretical part of the thesis focusses on the use of modern bioanalytical methods to determine heavy-metal cytotoxicity in human suspension cell lines. The experimental part is devoted to the preparation and cultivation of HL-60, Jurkat and Raji cell lines. An additional topic of the thesis is to describe the cytotoxic effects of CdCl2, HgCl2 and cisplatin on the Jurkat cell line. The determination of intracellular glutathione concentration and dehydrogenase activity of Jurkat cells was used to detect functional changes in the cell line tested. The selected tests were evaluated in the time range of 1-6 h and 24 h. As a conclusion, in the diploma thesis, all measured results are described and subsequently discussed.
