Advanced methods for characterisation of immunointeractions

Abstract:

Imunostanovení představují nejrozšířenější bioanalytickou metodu využívanou v mnoha oblastech od biologické bezpečnosti, přes ochranu životního prostředí po potravinářský průmysl. Imunostanovení jsou základem pro rozšíření použitelnosti elektrochemických senzorů na bio- a imunosenzory. Jedinečná kombinace těchto technologií dala vzniknout vysoce účinné možnosti detekce analytů. Pro další zvýšení citlivosti a snížení časové náročnosti lze v biosenzorech využít různé typy nanomateriálů, které nacházejí uplatnění především jako značky s katalytickými a optickými vlastnostmi. Tímto způsobem mohou být zlepšeny parametry i dalších technik využívajících protilátky, jako je např. imunohistochemie a imunocytochemie. Experimentální část práce představují přiložené recenzované publikace z impaktovaných časopisů. Pozornost byla zaměřená na použití nanočástic pruské modři pro detekci bakterie Salmonella Typhimurium a lidského sérového albuminu s limitem detekce (LOD) 6,0 × 103 CFU ml- 1, resp. 1,2 ng ml-1. Foton-upkonverzní nanočástice byly použity v imunostanoveních pro detekci Melissococcus plutonius s LOD 340 CFU ml-1 a v imunocytochemických testech pro vizualizaci HER2 receptorů s vysokým poměrem signálu k pozadí 319. Vyvinutý amperometrický imunosenzor pak umožňuje určit přítomnost patogenu Melissococcus plutonius s LOD 6,6 × 104 CFU ml-1.

Abstract:

Immunoassays represent the most widely used bioanalytical method for analyte detection and determination in various areas from biosecurity, through environmental protection, to food industry. Furthermore, the immunoassay principles serve as the basis for extending the applicability of electrochemical and optical sensors to bio- and immunosensing. The unique combination of these technologies gave rise to highly efficient methods for analyte detection. For even greater sensitivity and to lower the time consumption, biosensors can be modified using various types of nanomaterials. These can be used, for example, as labels with catalytic or optical properties. In this way, performance of various other techniques based on antibodies, such as immunohistochemistry and immunocytochemistry, can be enhanced. The experimental part of the dissertation is represented by the attached peer-reviewed publications in impacted journals. The work focused on the use of Prussian blue nanoparticles for the detection of Salmonella Typhimurium and human serum albumin with limit of detection (LOD) of 6.0 × 103 CFU mL−1 and 1.2 ng mL−1, respectively. Photon-upconversion nanoparticles were used in the immunosorbent assays for the detection of Melissococcus plutonius with LOD of 340 CFU mL−1 and in the immunocytochemical assays for visualization of HER2 receptors with an unsurpassed signal-to-background ratio of 319. The developed amperometric immunosensor was able to determine presence of the pathogen Melissococcus plutonius with LOD 6.6 × 104 CFU mL−1.
