Stilbene phytoalexins of grapevine

Abstract:

Stilbenové fytoalexiny jsou biologicky aktivní sloučeniny s nízkou molekulovou hmotností. Mohou být přítomny jako stálá obrana rostlinné buňky, převážně jsou však syntetizovány de novo po exposici stresovému faktoru, například při napadení patogenem. Rostlinné receptory v tomto případě rozpoznají látky zvané elicitory, čímž je spuštěna obranná reakce. Vhodné elicitory by mohly být používány k imunizaci rostlin cestou, která je šetrná k životnímu prostředí. Bylo by tak možno omezit jejich ošetřování syntetickými látkami, jako jsou pesticidy či fungicidy. V této diplomové práci byla zkoumána produkce stilbenů u buněčných suspensí Vitis vinifera cv. Gamay v reakci na působení elicitorů ergosterolu a oligogalakturonidů a v reakci na působení houbového patogenu Botrytis cinerea. Detekce probíhala pomocí vysokoúčinné kapalinové chromatografie s reversní fází. Byly detekovány extracelulární i intracelulární stilbeny neznámého složení a rovněž strukturně známý trans-resveratrol a piceid. Antifugální účinky stilbenů byly ověřeny inhibicí růstu mycelia Botrytis cinerea.

Abstract:

Stilbene phytoalexins are biologically active compounds having low molecular weight. They may be present in plant cells as a permanent defense, however, they are mostly synthesized de novo after exposure to stressful factors, such as pathogen attack. In this case, plant receptors recognize substances called elicitors, which are causing defensive reaction. Suitable elicitors could be used to immunize plants in environmentally friendly way. Thus it would be possible to avoid their treatment with synthetic substances, such as pesticides and fungicides. This diploma thesis investigated stilbene production in cell suspension of Vitis vinifera cv. Gamay in response to elicitation of ergosterol and oligogalacturonides and in response to the action of the fungal pathogen Botrytis cinerea. Detection was carried out using high performance liquid chromatography with reverse phase. Extracellular and intracellular stilbenes with yet unidentified composition were detected but also well-known structures of trans-resveratrol and piceid were found in our samples. Antifungal effects of stilbenes were verified by inhibiting the growth of Botrytis cinerea mycelia.
