Synthesis and Biological Assessment of Insecticides Based on Tacrine Scaffold

Abstract:

Diplomová práce se zabývá novými insekticidy na bázi takrinu jakožto duálními inhibitory enzymu acetylcholinesterasy (AChE). V rešeršní části práce jsou shrnuty vlastnosti a vývoj známých insekticidů, základní poznatky o nejčastěji se vyskytující se nemoci způsobené komárem - malárii, a v neposlední řadě informace o inhibitorech AChE, které představují experimentální podklad této práce. Hlavním výsledkem této diplomové práce je syntéza dvou sérií hybridních sloučenin, celkem 14 takrinových derivátů, u kterých byla dále stanovena inhibiční aktivita (vyjádřena hodnotami IC50) k lidské, muší a komáří AChE. Meziprodukty a cílové sloučeniny byly charakterizovány pomocí 1H NMR, 13C NMR, hmotnostní spektrometrií a elementární analýzou.

Abstract:

This diploma thesis deals with new insecticides based on tacrine as dual inhibitors of acetylcholinesterase enzyme (AChE). The research part of the thesis summarizes the properties and development of known insecticides, basic knowledge of the most frequently occurring diseases caused by mosquito - malaria, and last but not least information about inhibitors AChE, which represent the experimental basis of this work. The main result of this diploma thesis is the synthesis of two series of hybrid compounds, a total of 14 tacrine derivatives, in which the inhibitory activity (expressed by IC50 values) for human, fly and mosquito AChE was determined. Intermediates and target compounds were characterized by 1H NMR, 13C NMR, mass spectrometry and elemental analysis.
