Assessment of properties of fungal pathogens affecting the health status of beechnuts in vitro conditions

Abstract:

Tato bakalářská práce se zabývá hodnocením vlastností 10 vybraných houbových patogenů, které ovlivňují zdravotní stav bukvic. Veškeré testování probíhalo v podmínkách in vitro. Mezi vybrané patogeny patří druhy Mucor hiemalis, Apiognomonia errabunda, Diaporthe eres, Trichothecium roseum, Epicoccum nigrum, Rhizoctonia solani a Sclerotinia pseudotuberosa. Dále jsou to druhy rodu Alternaria sp., Fusarium sp. a Penicillium sp. Většina izolátů houbových patogenů byla získávána z bukvic buku lesního (Fagus sylvatica), druh R. solani byla získána z hlízy bramboru a druh S. pseudotuberosa ze žaludu dubu zimního (Quercus petraea). Testování probíhalo ve Fytopatologické a mykologické laboratoři Ústavu ochrany lesů a myslivosti. V rámci testování byla řešena rychlost růstu houbových patogenů a jejich růstové chování v teplotách 0 °C, 5 °C, 10 °C, 15 °C, 20 °C, 25 °C, 30 °C, 35 °C, 40 °C. Později byly přidány tři mrazové teploty -5 °C, -10 °C a -20°C. Rychlosti růstu jednotlivých druhů houbových patogenů byly pravidelně kontrolovány a zaznamenávány. Vyhodnocení proběhlo v programu QuickPHOTO MICRO, výsledná data se vyhodnotila v programu Statistica, výstupem jsou grafy z jednotlivých dnů měření.

Abstract:

This bachelor thesis deals with the evaluation of the properties of 10 selected fungal pathogens that affect the health status of beechnuts. All testing was carried out under in vitro conditions. The selected pathogens include the species Mucor hiemalis, Apiognomonia errabunda, Diaporthe eres, Trichothecium roseum, Epicoccum nigrum, Rhizoctonia solani and Sclerotinia pseudotuberosa. In addition, species of the genera Alternaria sp., Fusarium sp. and Penicillium sp. Most of the fungal pathogen isolates were obtained from beechnuts of Fagus sylvatica, R. solani was obtained from potato tubers and S. pseudotuberosa from the acorn of Quercus petraea. Testing was carried out in the phytopathological and mycological laboratory of the Institute of Forest Protection and Hunting. Testing focused on the growth rate of fungal pathogens and their growth behaviour at temperatures of 0 °C, 5 °C, 10 °C, 15 °C, 20 °C, 25 °C, 30 °C, 35 °C, 40 °C. Later, three freezing temperatures of -5 °C, -10 °C and -20 °C were added. The growth rates of each fungal pathogen species were regularly monitored and recorded. The evaluation was carried out in QuickPHOTO MICRO, the resulting data were evaluated in Statistica, and the graphs from each day of measurements are the output.
