Signaling pathways in breast cancer metastasis and their investigation using targeted proteomics

Abstract:

Tato bakalářská práce se zabývá vybranými signálními drahami ovlivňující metastatický proces u karcinomu prsu. Jejich studium je důležité pro rozvoj cílené terapie. Jedná se o signální dráhy transformujícího růstového faktoru-β (TGF-β), mitogenem aktivovaných proteinkináz (MAPK), „wingless/integrated“ (Wnt), Notch a cadherin-catenin signalizaci. Teoretická část popisuje jednotlivé signalizace a zabývá se významem klíčových proteinů v metastazování. Experimentální část se následně věnuje identifikaci a kvantifikaci těchto proteinů prostřednictvím metody SWATH s cílem zjistit, které dráhy a proteiny je možné touto metodou v nádorových tkáních sledovat. Z celkového počtu 58 proteinů se podařilo identifikovat a zároveň kvantifikovat 12 a prokázalo se tak, že některé dráhy a proteiny lze sledovat v komparativních experimentech. Toto zjištění je hlavním přínosem práce.

Abstract:

This thesis focuses on selected signaling pathways attecting the metastatic process in breast cancer. Their study is important for the development of targeted therapies. These signaling pathways are transforming growth factor-β (TGF-β), mitogen-activated protein kinase (MAPK), wingless/integrated (Wnt), Notch and cadherin-catenin signaling. Theoretical part describes the individual signalings and highlights the most important proteins in metastasis. Subsequently, the experimental part focuses on the identification and quantification of these proteins with the SWATH method to determine which pathways and proteins are possible to observe in the tumor tissues using this method. We managed to identify and quantify 12 proteins, out of the number of 58. It demonstrated that some pathways and proteins can be monitored in comparative experiments. This observation is a major contribution of this thesis.
