Denaturation and aggregation states of insulin in its reduced and oxidized forms

Abstract:

The presented bachelor thesis deals with the study of various structural forms of recombinant human insulin (Human Insulin, HI) in aqueous buffered solutions and the reflection of these forms into its electrochemical behaviour. The approach combining spectral (circular dichroism, CD) and electrochemical (cyclic voltammetry, CV) experiments together with the Beta Structure Selection (BeStSel) numerical algorithm confirmed the correlation of structural changes (denaturation and aggregation states) and HI oxidation signals. Electrochemical experiments on screen printing graphite electrodes modified with bismuth nanoparticles have shown that tyrosine is responsible for the HI oxidation signal, which in this way establishes itself as an electroactive marker of HI conformational changes. In general, the insulin molecule contains a total of four tyrosines, and the extent of conformational changes can be judged by their position and their accessibility to the electrode surface. The dependences of the spectral and electrochemical outputs on various experimental conditions, such as pH, buffer composition, temperature, HI concentration, the presence of chlorides or denaturing agents, were revealed and discussed. It should be noted that insulin, as a relatively small polypeptide, is very flexible and very easily subject to the formation of oligomeric formations, especially dimer and hexamer. That is why the effort to accurately describe the molecular structures of insulin in various settings can be considered a very important step forward, both in terms of the development of the insulin sensor and in terms of possible treatment of diabetes mellitus (DM).

Abstract:

Predkladaná bakalárska práca sa zaoberá štúdiom rôznych štruktúrnych foriem rekombinantného ľudského inzulínu (Human Insulin, HI) vo vodných pufrovaných roztokoch a reflexií týchto foriem do jeho elektrochemického správania. Prístup kombinujúci experiment spektrálny (cirkulárny dichroismus, CD) a elektrochemický (cyklická voltametria, CV) spolu s využitím numerického algoritmu Beta Structure Selection (BeStSel) potvrdil koreláciu štruktúrnych zmien (denaturačných a agregačných stavov) a oxidačných signálov HI. Elektrochemické experimenty na sieťotlačových grafitových elektródach modifikovaných nanočasticami bizmutu ukázali, že za oxidačný signál HI je zodpovedný tyrozín, ktorý sa týmto spôsobom etabluje do postavenia elektroaktívneho ukazovateľa (marker) konformačných zmien HI. Všeobecne, molekula inzulínu obsahuje celkom štyri tyrozíny a podľa ich pozície a ich prístupnosti k elektródovému povrchu je možné posudzovať rozsah konformačných zmien. Boli odhalené a diskutované závislosti spektrálnych a elektrochemických výstupov na rôznych experimentálnych podmienkach, ako je pH, zloženie pufru, teplota, koncentrácia HI, prítomnosť chloridov alebo denaturačných činidiel. Je potrebné poznamenať, že inzulín, ako relatívne malý polypeptid, je veľmi flexibilný a veľmi ľahko podlieha tvorbe oligomerných formácií, najmä diméru a hexaméru. Práve preto možno považovať snahu o presný popis molekulových štruktúr inzulínu v rôznych prostrediach za veľmi významný krok vpred, a to ako z hľadiska vývoja inzulínového senzora, tak aj z hľadiska prípadnej liečby diabetes mellitus (DM).
