Insect immune and behavioural reactions under stress condition

Abstract:

Všechny živé organismy jsou během života ovlivňovány mnohými stresovými faktory, a aby přežily, musejí si proti nim vyvinout behaviorální, fyziologické a imunitní reakce. Úspěšná obrana proti těmto negativním vlivům vyžaduje přesouvání dostupných zdrojů organismu tak, aby bylo dosaženo selektivní výhody. Pro odhalení a pochopení základní mechaniky stresové reakce jsou na savcích uskutečňovány rozsáhlé studie dané problematiky. Nicméně studium stresových reakcí může být prováděno i na hmyzích modelových organismech, jelikož sdílí mnoho podobných stresových reakcí i řídicích mechanismů se savci. Navíc hmyzí modelové organismy mají oproti savcům i další výhody, jako kratší životní cyklus, snadnější manipulaci či nižší legislativní nároky nutné pro práci s bezobratlými. Ve své disertační práci se zabývám vlivem různých ekologických, chemických a biologických stresorů na behaviorální fyziologickou a imunitní reakci hmyzu. Zimní sezóna jako jeden z negativních ekologických stresorů zvýšila antimikrobiální aktivitu a koncentraci proteinů v hemolymfě včely medonosné (Apis mellifera), avšak vliv na ostatní živiny či na buněčnou část imunitních reakcí potvrzen nebyl. Na druhou stranu u zavíječe voskového (Galleria mellonella) došlo po podání dusičnanu draselného ke změně právě v buněčné složce imunity, konkrétně ke snížení schopnosti hemocytů bojovat s infekcí, přestože celkový počet hemocytů zůstal srovnatelný s kontrolami. Chemické stresory neovlivňují pouze fyziologické a imunitní parametry, ale také ty behaviorální. Larvy G. mellonella trpěly po podání kofeinu reverzibilní odeznívající po 48 hodinách. Opačného efektu, tedy zvýšení pohybové aktivity, bylo dosaženo po podání adipokinetického hormonu larvám octomilky obecné (Drosophila melanogaster). Změna chování však nemusí být pouze důsledkem při samotném působení stresoru, ale může být také ochranou před jeho působením. Například larvy D. melanogaster se dokáží záměrně vyhýbat oblastem s entomopatogenními hlísticemi, a tak snížit riziko infekce. Celková stresová odpověď tedy sestává z mnoha reakcí dílčích, které je nutno podrobně popsat. Jedině tak budeme schopni dopad stresorů na organismus plně pochopit.

Abstract:

Every animal species is influenced by various stressors during its life, and in order to survive, organisms had to evolve behavioural, physiological, and immune responses to counteract these negative factors. To successfully defend against negative stressors, animals have to reallocate available resources as the risks shift to gain a selective advantage. Stress reactions are extensively studied in mammals to understand the basic mechanisms and reveal the underlying molecular principles. However, stress responses can also be studied on insect model organisms due to many similarities in their stress reactions, which are driven by many analogous neurohormones, biogenic amines, and response pathways. Moreover, insect model organisms provide many more benefits, like short life cycle, easier handling, and the absence of ethical issues. We studied effects of selected ecological, chemical, and biological stressors on the “fight-or-flight” behavioural response, physiological parameters like nutrient concentrations, and humoral and cellular immune reactions. The winter season, a combination of several negative ecological stressors, increases antimicrobial activity in Apis mellifera as well as protein concentrations in the haemolymph. On the other hand, it has no effect on other nutrient levels or cellular immunity. Nevertheless, reduced cellular immune reactions were proved in Galleria mellonella larvae after the administration of potassium nitrate as chemical stressor, although the total number of haemocytes remained similar to the controls. Chemical stressors are able to affect not only physiological and immune parameters but also behavioural responses. G. mellonella larvae suffered reversible paralysis and impaired development after artificial administration of caffeine. Entirely opposite effect was observed in Drosophila melanogaster larvae, whose locomotor activity increased after administration of adipokinetic hormone which is one of the insect stress hormones. However, such behaviour does not have to be only the consequence of a stress reaction but it can serve a protective function. For example, D. melanogaster larvae deliberately avoid contact with entomopathogenic nematodes, which prevents them from potentially contracting an infection. Stress response therefore consists of many different reactions, which have yet to be accurately described to fully understand the effects of stressors on organisms.
