Determination of an alternative toxicological in vitro eye irritation test by HET-CAM method

Abstract:

Diplomová práce se zabývá alternativními metodami, které se používají pro odhalení potenciálu látek způsobit poškození nebo podráždění oka. V experimentální části práce je na vybrané skupině látek ověřen nový způsob vyhodnocení testu oční dráždivosti na chorioalantoidní membráně kuřecího embrya pomocí barviva Trypan blue (Trypanové modři). Do experimentů byly použity další dvě alternativní metody HET-CAM a BCOP. Testování probíhalo na chemikáliích se známým in vivo účinkem. Výsledky testů byly mezi sebou porovnány. Závěrem byla doporučena kombinace dvou alternativních metod dosahující 82,4% shody s in vivo výsledky.

Abstract:

The diploma thesis deals with alternative methods which are used to reveal the potential of substances to cause damage or irritation to the eye. In the experimental part of the work, a new method of evaluating the eye irritation test on the chorioallantoic membrane of the chicken embryo using the dye Trypan blue is verified on a selected group of substances. Two other established alternative methods, HET-CAM and BCOP, were also tested. Testing was performed on chemicals with known in vivo effects. The test results were compared with each other. In conclusion, a combination of two alternative methods was recommended, achieving 82.4% agreement with the in vivo results.
