Telomeres in plants with modified genomes

Abstract:

Moss Physcomitrium patens is the important plant model in telomere biology. Telomeres are nucleoprotein complexes located at the ends of linear eukaryotic chromosomes that are crucial for the genome integrity. Length of telomeres is maintained by the enzyme telomerase. In case of a loss of function of any telomerase subunit, alternative lengthening of telomeres by a mechanism of homologous recombination can be activated. The aim of the study was to verify the viability of the P. patens line with a mutation in both alleles of the RAD51 gene, encoding an important protein of homologous recombination, and a reduced level of TERT, a catalytic subunit of telomerase. The theoretical part of the bachelor thesis contains information about the model organism Physcomitrium patens. Furthermore, various types of DNA damage and their repair mechanisms are described. In the experimental part, the presence of mutant RAD51-1, RAD51-2 alleles was verified and the level of TERT transcript was analysed in Physcomitrium patens mutants with the presumptive genotype rad51-1,2/tert. In the bachelor thesis, a significant reduction in the level of TERT transcript in the mutant line was confirmed. Analysis of the RAD51 gene alleles revealed the presence of the standard RAD51-1 allele and the mutant RAD51-2 allele, which was not expected. Analysis of the original rad51-1,2 mutant line revealed the presence of both, standard and mutant RAD51-1 alleles; the RAD51-2 allele was present only in a mutant form. Since only cells with the standard RAD51-1 allele were present after a reduction of TERT transcript level, can be assumed that we are unable to obtain a viable culture of P. patens with mutations in both RAD51-1 and RAD51-2 alleles and significantly reduced TERT level.

Abstract:

Mach Physcomitrium patens je jedným z významných rastlinných modelov v biológii telomér. Teloméry sú nukleoproteínové komplexy lokalizované na koncoch lineárnych eukaryotických chromozómov, ktoré sú dôležité pre zachovanie integrity genómu. Stabilná dĺžka telomér je udržiavaná enzýmom telomerázou. Pri poruche funkcie ktorejkoľvek jej podjednotky môže dôjsť k aktivácii alternatívneho predlžovania telomér mechanizmom homologickej rekombinácie. Cieľom práce bolo overenie životaschopnosti línie P. patens s mutáciou v oboch alelách génu kódujúcich RAD51, ktorý je dôležitým proteínom homologickej rekombinácie, a zníženou hladinou TERT, katalytickej telomerázovej podjednotky. V teoretickej časti bakalárskej práce sú uvedené informácie o modelovom organizme Physcomitrium patens. Ďalej sa práca zaoberá rôznymi druhmi poškodení DNA a ich opravami. V experimentálnej časti bola overená prítomnosť mutantných foriem alel RAD51-1, RAD51-2 a analyzovaná hladina transkriptu TERT v mutantoch Physcomitrium patens s predpokladaným genotypom rad51-1,2/tert. V bakalárskej práci bolo potvrdené výrazné zníženie hladiny transkriptu TERT v mutantnej línii. Analýzou alel génu RAD51 bola zistená prítomnosť štandardnej alely RAD51-1 a mutantnej alely RAD51-2, čo nezodpovedalo očakávanému genotypu. Analýzou pôvodnej mutantnej línie rad51-1,2 bola zistená prítomnosť ako štandardnej, tak mutantnej alely RAD51-1; alela RAD51-2 bola prítomná iba ako mutantná. Nakoľko po znížení hladiny transkriptu TERT boli prítomné len bunky so štandardnou alelou RAD51-1, je možné predpokladať, že nie sme schopní získať viabilnú kultúru P. patens s mutáciou oboch alel RAD51-1, RAD51-2 a výrazne zníženou hladinou TERT.
