Effect of CCD inhibitors on results determination of multiplex methods ISAC and Alex

Abstract:

Tato bakalářská práce se zabývá zhodnocením vlivu inhibitoru zkříženě reagujících sacharidových determinant (CCD) u metody ALEX na výsledky stanovení hladin specifických IgE protilátek proti různým alergenovým molekulám a srovnáním těchto výsledků s metodou ISAC. Zhodnocení vlivu CCD inhibice jsem prováděla pomocí porovnání četnosti pozitivních výsledků u shodných nativních alergenových molekul mezi metodami ISAC a ALEX. Při měření metodou ISAC jsem získala celkem 140 pozitivních výsledků a při měření metodou ALEX jsem naměřila celkem 133 pozitivních hodnot. Po vyhodnocení získaných výsledků jsem prokázala účinnou CCD inhibici, protože počet pozitivit při měření metodou ISAC byl vyšší než při měření metodou ALEX. Z tohoto důvodu je lepší využívat u vzorků sér, které mají vysoké riziko zkřížené reaktivity, vyšetření pomocí metody ALEX, která díky CCD inhibitoru snižuje množství falešně pozitivních výsledků.

Abstract:

This bachelor thesis deals with the evaluation of the effect of an inhibitor of cross-reactive carbohydrate determinants (CCD) in the ALEX method on the results of determining the levels of specific IgE antibodies against various allergen molecules and comparing these results with the ISAC method. I evaluated the effect of CCD inhibition by comparing the frequency of positive results for identical native allergen molecules between the ISAC and ALEX methods. When measured by the ISAC method, I obtained a total of 140 positive results, and when measured by the ALEX method, I measured a total of 133 positive values. After evaluating the obtained results, I demonstrated effective CCD inhibition, because the number of positives in the ISAC measurement was higher than in the ALEX measurement. For this reason, it is better to use ALEX testing of serum samples that have a high risk of cross-reactivity, which reduces the number of false-positive results due to a CCD inhibitor.
