Dopamine and its Use in the Thiol-modified Hyaluronan Based Hydrogels

Abstract:

V diplomové práci je představena syntéza crosslinkeru (polydopaminu) pro přípravu hydrogelů na bázi thiolovaného hyaluronanu (HA-SH), ale také syntéza hyaluronanu konjugovaného dopaminem (HA-DA) jako dalšího způsobu využití dopaminu při vývoji inovativních materiálů pro biomedicínské aplikace. Dílčím cílem práce bylo zhodnotit vliv parametrů přípravy polydopaminu (PAD) ve formě částic i trubiček, optimalizovat podmínky syntézy a provést charakterizaci PAD, následně připravit hydrogely na bázi syntetizovaných hyaluronových derivátů a PAD částic/trubiček a provést jejich charakterizaci pomocí dostupných metod. Úspěšné navázání thiolových a katecholových skupin na řetězec hyaluronanu byla potvrzena pomocí NMR spektroskopie. Velikost polymerních klubek HA-SH a HA-DA, a také velikost PAD byla stanovena metodou DLS. Připravené hydrogely byly charakterizovány reologickým měřením z pohledu doby gelace, tuhosti hydrogelů a reverzibility reologického chování po uvolnění napětí. Morfologie PAD částic a trubiček a morfologie a porozita hydrogelů byla studována pomocí SEM. V neposlední řadě byla stanovena botnací kapacita hydrogelů. Na základě měření NMR byla potvrzena úspěšnost navázání thiolových, resp. katecholových, skupin na řetězci HA(Na) se stupněm substituce 9 %, resp. 29 %. Byly připraveny PAD částice s úzkou distribucí velikosti, jejichž velikost byla a 627 ? 10 nm. Nejvyšší tuhost vykazovaly hydrogely síťované PAD ve formě trubiček. Dobrou reverzibilitu vykazovaly hydrogely připravené z thiolovaného hyaluronanu HA-SH (z 64 Pa (1%) na 58 Pa (100%) a zpět na 68 Pa (1%)) a hydrogely připravené z hyaluronanu konjugovaného dopaminem HA-DA (ze 45 Pa (1%) na 40 Pa (100%) a zpět na 44 Pa (1%)). Nejnižší reverzibilitu vykazovaly hydrogely síťované PAD (ve formě částic i trubiček, a to pouze 84, resp. 91 %). Nejvyšší porozita byla pozorována u hydrogelů na bázi HA-DA, v jejichž struktuře byly detekovány póry o veliskoti (> 100 m).

Abstract:

The synthesis of a crosslinker (polydopamine) for the preparation of hydrogels based on thiolated hyaluronan (HA-SH), but also the synthesis of dopamine-conjugated hyaluronan (HA-DA) as another way to use dopamine in the development of innovative materials for biomedical applications is presented in the thesis. The sub-objectives of this work were to evaluate the influence of polydopamine (PAD) preparation parameters in the form of particles and tubes, to optimize the synthesis conditions and to characterize PAD, and then to prepare hydrogels based on the synthesized hyaluronan derivatives and PAD particles/tubes and to characterize them using available methods. The successful attachment of thiol and catechol groups to the hyaluronan chain was confirmed by NMR spectroscopy. The size of the polymeric HA-SH and HA-DA clubs, as well as the size of the PAD, was determined by DLS. The prepared hydrogels were characterized by rheological measurements in terms of gelation time, hydrogel stiffness and reversibility of rheological behavior after stress release. The morphology of PAD particles and tubes and the morphology and porosity of hydrogels were studied by SEM. Finally, the botnation capacity of the hydrogels was determined. NMR measurements were used to confirm the successful attachment of thiol and catechol groups on the HA(Na) chain with a degree of substitution of 9% and 29%, respectively. PAD particles with a narrow size distribution were prepared with a size of 627 ? 10 nm. The highest stiffness was exhibited by the crosslinked PAD hydrogels in the form of tubes. Hydrogels prepared from thiolated HA-SH hyaluronan (from 64 Pa (1%) to 58 Pa (100%) and back to 68 Pa (1%)) and hydrogels prepared from dopamine-conjugated HA-DA hyaluronan (from 45 Pa (1%) to 40 Pa (100%) and back to 44 Pa (1%)) showed good reversibility. The hydrogels cross-linked with PAD (in the form of particles and tubes, only 84 and 91%, respectively) showed the lowest reversibility. The highest porosity was observed for HA-DA-based hydrogels, in which pores of the size (> 100 m) were detected in the structure.
