Microwave-assisted synthesis of 2,6,9-trisubstituted purines

Abstract:

Možná biologická aktivita purinů motivuje mnohé vědecké týmy k výzkumu možností syntéz jejich 2, 6, 8 a 9 substituovaných derivátů. Tato diplomová práce popisuje tříkrokovou syntézu 2,6,9-trisubstituovaných purinů s doposud nepopsanou strukturou. Samotná syntéza sestávala z alkylace komerčně dostupného 2,6-dichlor-9H-purinu 2-jodpropanem, následovanou mikrovlnami asistovanou SNAr atomu chloru v poloze 6 alkylovaného purinu, a v posledním kroku další mikrovlnami asistovanou SNAr atomu chloru v poloze 2 purinových derivátů, která se ukázala být relativně problematickou. Připravené sloučeniny byly charakterizovány pomocí běžně používaných metod strukturní analýzy.

Abstract:

The possible biological activity of purines motivates many research groups to investigate the possibility of synthesizing their 2, 6, 8 and 9 substituted derivatives. This diploma thesis describes a three-step synthesis of 2,6,9-trisubstituted purines with an undescribed structure. The synthesis itself consisted of the alkylation of commercially available 2,6-dichloro-9H-purine with 2-iodopropane, followed by microwave-assisted SNAr of the chlorine atom in position 6 of the alkylated purine, and in the last step another microwave-assisted SNAr of chlorine in position 2 of the purine derivatives, proved to be relatively problematic. The prepared compounds were characterized using commonly used structural analysis methods.
