Use of hydrodistillation and steam distillation for extraction of volatile substances from plant material

Abstract:

Předmětem diplomové práce jsou esenciální oleje. Teoretická část je zaměřena na chemické a biologické vlastnosti vybraných esenciálních olejů a jejich využití. Dále se práce zabývá izolací esenciálních olejů a jejich charakterizací plynovou chromatografií s hmotnostní detekcí (GC-MS) a plynovou chromatografií s plamenovým ionizačním detektorem (GC-FID). Experimentální část práce se zabývá zejména hydrodestilací a parní destilací v klasickém uspořádání a vlivem těchto extrakčních technik na složení esenciálních olejů. Esenciální oleje jsou analyzovány GC-MS a GC-FID. Dále práce sleduje antimikrobiální a antioxidační vlastnosti esenciálních olejů. Práce je zaměřena na 4 usušené matrice květy levandule lékařské, listy vavřínu ušlechtilého, květy hřebíčkovce kořenného a semena fenyklu obecného. Antioxidační účinky esenciálních olejů byly hodnoceny metdou s difenylpikryl hydrazylovým radikálem. Antimikrobiální účinky esenciálních olejů byly hodnoceny metodou diskového difúzního testu (DDT) ve srovnání s několika vybranými antibiotiky. V případě kvalitativní metody DDT se citlivost mikroorganismu projeví inhibiční zónou růstu. Testy byly prováděny na čtyřech vybraných druzích mikroorganismů Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa a Candida albicans.

Abstract:

The subject of the diploma thesis is essential oils. The theoretical part is focused on the chemical and biological properties of selected essential oils and their ussage. In addition, the thesis deals with the isolation of essential oils and their characterization by gas chromatohraphy with mass spektrometry detection (GC-MS) and gas chromatography with flame ionization detection (GC-FID). The experimental part deals mainly with hydrodesilation and steam distillation in a classical setup and the influence of these extraction techniques for the composition of essential oils. Essential oils are analyzed by GC-MS and GC-FID. Furthermore, the thesis follows the antimicrobial and antioxidant properties of essential oils. The work is focused on 4 dried matrices - lavender flowers, laurel leaves, clove flowers and fennel seeds. The antioxidant properties of essential oils were evaluated by diphenylpicrylhydrazyl radical method. The antimicrobial effects of essential oils were evaluated by a disk diffusion test (DDT) method compared to several selected antibiotics. In the case of the DDT qualitative method, the susceptibility of the microorganism to the growth inhibition zone is demonstrated. The tests were conducted on four selected microorganisms - Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa and Candida albicans.
