Abstract:

Podstatou diplomové práce bylo studium stereoselektivních vlastností vybraných lipáz při katalýze hydrolýzy triacylglycerolů na základě analýzy získaných produktů izomerů monoacylglycerolů a diacylglycerolů s využitím chirální superkritické fluidní chromatografie s hmotnostně spektrometrickou detekcí. Pro identifikaci produktů hydrolýzy byly využity předem syntetizované enantiomerně čisté standardy monoacylglycerolů a diacylglycerolů. Stereoselektivita vybraných lipáz byla studována na základě poměrů v zastoupení enantiomerů ve vzorku po hydrolýze. Součástí diplomové práce je také literární rešerše na téma analýzy vzorků lipidů, zejména monoacylglycerolů a diacylglycerolů.

Abstract:

The goal of the diploma thesis was the study of the stereoselective properties of selected lipases during the catalysis of the hydrolysis of triacylglycerols based on the analysis of the obtained products of isomers of monoacylglycerols and diacylglycerols using chiral supercritical fluid chromatography with mass spectrometric detection. Pre-synthesized enantiomerically pure standards of monoacylglycerols and diacylglycerols were used to identify the hydrolysis products. The stereoselectivity of the selected lipases was studied based on the ratio of enantiomers in the sample after hydrolysis. The diploma thesis also includes a literature search on the topic of lipid sample analysis, especially monoacylglycerols and diacylglycerols.
